复合RT-PCR检测猪繁殖与呼吸综合征和猪瘟及猪流感等3种病毒方法的建立

摘要

为了快速准确地对猪繁殖与呼吸综合征病毒（PRRSV）、猪瘟病毒（CSFV）和猪流感病毒（SIV）进行诊断与鉴别诊断，特进行试验。根据GenBank上已发表的PRRSV的ORF7基因序列、CSFV的C基因序列和SIV的M基因序列，设计合成了3对特异引物，开展RT-PCR检测方法的研究。分别建立了PRRSV、CSFV和SIV的单项RT-PCR诊断方法，并最终建立了PRRSV、CSFV和SIV的复合RT-PCR诊断方法，可分别及同时扩增PRRSV 430 bp、CSFV 775 bp和SIV 225 bp的特异性片段。试验证明该方法适用于PRRSV、CSFV和SIV的诊断及鉴别诊断。%In order to diagnose and differential diagnose for PRRSV,CSFV and SIV,this experiment was carried out.Based on the ORF7 gene sequences of PRRSV,C gene sequences of CSFV and M gene sequences of SIV obtained from GenBank,three pairs of primers were designed and used for RT-PCR.Single RT-PCR assays for diagnosing PRRSV,CSFV and SIV were established,the multiplex RT-PCR was also developed in the end,the fragments of 430 bp for PRRSV,775 bp for CSFV and 225 bp for SIV could be amplified in single RT-PCR and a multiplex RT-PCR assay.According to the test,the RT-PCR assay was suit to diagnose and differential diagnose for PRRSV,CSFV and SIV.
