摘要

Objective To examine the role of sphingosine 1-phosphate ( S1 P) signals in the regulation of rat vascular smooth muscle cells migration, and to offer the new ideas for tumor angiogenesis and cardiovascular disease. Methods Rat vascular smooth muscle cells were isolated and characterized by expression of α-smooth muscle(α-SM) actin. The hypoxia models were induced by 1% O2 incubators and CoCl2. The expression patterns of S1P receptor mRNAs in rat vascular smooth muscle cells were analyzed using RT-PCR analysis. The Transwells were used for the role of S1P signals in the regulation of rat vascular smooth muscle cells migration, that was confirmed by the S1P receptor antagonist. Results and Conclusion SPK1 and the S1P receptor, rS1P1, rS1 P2 and rS1 P3 were expressed in rat vascular smooth muscle cells. Hypoxia induced by 1% O2 and CoCl2 could upregulate SPK1 expression and activity of rat vascular smooth muscle cells. S1P promotes the migration of rat vascular smooth muscle cells through S1P receptor subtypes rS1P2.%目的 探讨1-磷酸鞘氨醇(sphingosine 1-phosphate,S1P)对大鼠血管平滑肌细胞(rat vascular smooth muscle cells,rVSMc)迁移的作用机制,为肿瘤血管新生和心血管疾病发生机制的研究提供新的思路.方法 体外分离培养、鉴定rVSMc细胞,建立低氧培养箱和氯化钴诱导的细胞低氧模型,应用RT-PCR方法对rVSMc细胞的SIP受体表达水平进行检测,运用微孔隔离室穿越方法研究SIP对rVSMc细胞迁移的作用,并用S1P受体阻断刺加以证实.结果 与结论 rVSMc细胞表达SPK1和S1P受体rS1P1、rS1P2和rS1P3,低氧状态下rVSMc细胞SPK1的表达和活性均高于对照,S1P主要通过与rS1P2受体结合促进rVSMc细胞的迁移.
