Langendorff离体心脏灌注模型的制备及应用

摘要

The isolated heart perfusion model is set up by perfusion through the catheter intubated backward into aorta to coronary artery to supply myocardium nutrition to keep the isolated heart beating. It can be used in heart function study at organ level;or it can also be perfused with solution containing collagenase for preparation isolated myocytes used in the study of the myocyte function at cell or molecule level. The kind of animal selected for Langendorff perfusion is decided by research purpose and condition. To prepare an ideal Langendorff perfusion heart needs appropriate solution and perfusion pressure and temperature,skilled anaesthesia heart taking and intubating aorta and perfusing operation, ensuring good heart perfusion, which are essential to get functional isolated beating heart or myocytes for the study of heart physiology,pharmacology or clinical basic application. Although it has some limits compared with the heart in vivo,the preparation of isolated Langendorff perfusion heart is easy and economical with good repeatability,making it appropriate for the heart function research.%离体心脏灌注模型通过主动脉逆行插管灌注灌流液,经冠状动脉血管营养心肌,维持离体心脏持续搏动,可用于器官水平心脏功能研究;或灌注含胶原酶的溶液分离单个心肌细胞用于心肌细胞或分子功能研究.根据研究目标条件选用适宜动物,成功制备离体心脏Langendorff灌注模型,需要选用合宜的灌注液、控制灌注压力流量及温度,娴熟的麻醉取心、主动脉插管及灌注操作,保证离体心脏良好灌注,才能制得功能良好的离体搏动心脏或心肌细胞,用于心脏生理、药理、临床应用基础研究.其与在体心脏相比有一定局限性,但该模型制备简便经济,重复性好,适用于心脏功能学研究.
