Toll样受体4与移植器官缺血/再灌注损伤

     

摘要

Toll-like receptor 4(TLR4)is one of the most important receptors of innate immunity which is responsible for the initial response of the organism to pathogens and harmful stimulus. TLR4 participates in inflammatory response triggered by infectious stimuli, and recognizes endogenic ligands( heat shock protein, high mobility group protein B1 ,etc. )released from impaired cells via ischemia/reperfusion injury,which induces proinflammatory cytokines by TLR4/MyD88 signal pathway. And negtive regulation of TLR4 signaling inhibits transplant-induced ischemia/reperfusion injury in various animal experiment. So, it is promising to protect transplanted organ against ischemia/reperfusion injury via regulation of TLR4 signaling in clinical practice.%Toll样受体4(TLR4)是抵御病原体入侵和有害刺激的重要天然免疫受体之一.TLR4除了参与病原微生物触发的炎性反应外,还能识别缺血/再灌注损伤后释放的内源性配体,如热休克蛋白、高迁移率族蛋白B1等,经TLR4/髓样分化因子88等信号途径诱导多种炎性细胞因子的产生,参与移植器官缺血/再灌注损伤的炎性反应.实验表明,负性调节TLR4信号对移植器官缺血/再灌注损伤具有明显的保护作用.因此,针对TLR4信号的调节在移植器官保护方面具有一定的临床应用前景.
