嗜酸性粒细胞趋化因子和基质金属蛋白酶9在鼻息肉中的表达

     

摘要

目的:探讨嗜酸性粒细胞趋化因子(Eotaxin)和基质金属蛋白酶9(MMP-9)在息肉组织中的表达及其与嗜酸性粒细胞( Eos)活化和发病的关系。方法选择2010年7月至2013年7月在荆州市中心医院住院接受鼻息肉手术治疗的患者50例作为观察组,并选取35例鼻外伤及慢性鼻炎患者作为对照组。分别于鼻内镜下摘取观察组患者鼻内息肉组织和对照组患者鼻下甲鼻黏膜组织进行苏木精-伊红染色和免疫组织化学SP染色,观察Eotaxin和MMP-9的表达及Eos浸润情况。结果观察组患者中血清嗜酸粒细胞阳离子蛋白(ECP)水平显著高于对照组[(17.32±4.62)μg/L vs (3.28±1.22)μg/L],差异有统计学意义(P<0.01)。观察组患者Eotaxin阳性细胞数显著高于对照组[(47.25±13.18)个 vs (12.48±10.12)个],差异有统计学意义(P<0.01)。 MMP-9在非鼻息肉的正常损伤鼻内组织中呈弱阳性表达,而在观察组患者中MMP-9均呈强阳性表达,两组平均灰度值比较差异有统计学意义(P<0.01)。结论息肉组织中,Eotaxin和MMP-9可通过增加Eos浸润,引发组织的异常重塑,在鼻息肉存在和发展中起重要作用。%Objective To investigate the eosinophil chemotactic factor and matrix metalloproteinase-9 (MMP-9) expression in polyps with eosinophils extremely ( Eos) activation and morbidity relationship. Methods 50 cases with nasal polyps admitted to Jingzhou Central Hospital from July 2010 to July 2013 were chosen as observation group,35 cases patients with nasal trauma,chronic rhinitis causes nasal were cho-sen as control group. Endoscopic removal of the study were patients with nasal polyps group and a control group of patients under the nose A nasal tissue HE staining and immunohistochemical SP staining eosinophil chemotactic factor( eotaxin) and MMP-9 expression and eosinophil infiltration. Results The patients serum EPC level in the observation group was significantly higher than the control(17. 32 ± 4. 62) μg/L vs (3. 28 ±1.22) μg/L)(P<0.01). Eotaxin-positive cell counts in observation group was higher than the control [(47.25±13.18)个 vs(12.48±10.12)个](P<0.01).MMP-9inanon-normaldamagenasalpolyp tissue was weakly positive expression,whereas patients in the observation group,MMP-9 showed a strong ex-pression of the average gray value of the two groups reached statistically significant difference compared levels (P<0. 01). Conclusion Eosinophil chemotactic factor and matrix metalloproteinase-9 can increase eosin-ophil infiltration,causing the abnormal tissue remodeling,the existence and development of nasal polyps play an important role.
