毛细管电泳法测定罗布麻叶中芦丁、异槲皮苷和槲皮素的含量

         

摘要

Objective To establish a high -performance capillary electrophoretic (HPCE) method for simultaneous determination of contents of rutin, isoquercitrin, and quercetin in Apocynum venetum. Methods A bare fused — silica capillary (58.5 cm x 75 (j,m, effective length of 48. 5 cm) was used. The buffer was composed of 50 mmol/L Na2B4O7 ( pH 9. 60) . The running voltage was set at 20 kV with polarity from anode ( inlet) to cathode ( outlet) . The separation time was 20 min and temperature was set at 25 'C . Online detection was performed at 350 nm ( PDA). Results The three types of flavonoids were well separated in 25 minutes and linear relationship in the peak area was good. The linear range was 4.4—44, 2.8—28.2, and3.8—38 (j,g/ml, respectively. The correlation coefficient was 0. 9968, 0. 9995 , and 0. 9991, respectively. The average recovery was 97. 3% , 98. 4% , and 98. 3% , respectively. Conclusions This method is simple, rapid, accurate, and reliable, which can be used for content determination of flavonoids in Apocynum leaves.%目的 建立毛细管电泳法(capillary zone electrophoresis,CZE)同时分离测定罗布麻叶中的芦丁、异槲皮苷和槲皮素3种黄酮类化合物.方法 采用熔融石英毛细管(58.5 cm×75 μm),有效长度为48.5 cm(未涂层);缓冲体系为50 mmol/L Na2B4O7 (pH9.6+15%乙腈);分离电压 20 kV(+)→(-);电泳时间 20 min;电泳温度25 ℃;压力进样 0.5 psi×20 s;柱上检测 UV 350 nm(二极管阵列检测器).结果 3种黄酮类化合物在25 min内得到较好分离,芦丁、异槲皮苷、槲皮素的线性范围分别为4.4~44、2.82~28.2、3.8~38 μg/ml,相关系数为0.9968、0.9995、0.9991,与峰面积线性关系良好;样品平均回收率分别为97.3%、98.4%和98.3%.结论 本方法准确可靠,可用于罗布麻叶中黄酮类成分的含量测定.
