香鳞毛蕨提取物对红色毛癣菌的抑制作用研究

         

摘要

目的:研究香鳞毛蕨醇提取物对红色毛癣菌角鲨烯环氧化酶和β-(1,3)-D-葡聚糖合成酶的抑制作用,初步探讨其抗真菌作用机制。方法采用 ELISA测定角鲨烯环氧化酶活性,还原糖法测定β-(1,3)-D-葡聚糖合成酶活性。结果香鳞毛蕨提取物不同浓度组均能降低红色毛癣菌角鲨烯环氧化酶的活性,差异有统计学意义(P<0.05),随香鳞毛蕨提取物浓度增加,红色毛癣菌角鲨烯环氧化酶的活性逐渐降低,香鳞毛蕨提取物中浓度组和高浓度组能降低β-(1,3)-D-葡聚糖合成酶活性,差异有统计学意义(P=0.002,P=0.000)。结论香鳞毛蕨提取物抗真菌作用机制可能和角鲨烯环氧化酶和β-(1,3)-D-葡聚糖合成酶活性受到抑制有关。%Objective To study the inbibitional ef ect of alcohol extract of Dryopteris fragrans ( L.) Schot on squalene epoxidase and β- (1,3) - D - glucan synthase of Trichophyton rubrum and its antifungal mechanism. Methods Determination of squalene epoxidase activity by ELISA test,determination of β- (1,3) - D - glucan synthase activity by method of reducing sugar. Results Extract of Dryopteris fragrans(L.) Schot in dif erent concentration can reduce Trichophyton rubrum squalene epoxidase activity (with statistical significance,P<0.05),with the increasing concentration of the extract of Dryopteris fragrans (L.) Schot ,Trichophyton rubrum squalene epoxidase activity gradual y reduced,the extraction of Dryopteris fragrans(L.) Schot medium concentration group and high concentration group can decreaseβ-(1,3)-D-glucan synthase activity (statistical y significant dif erence,P=0.002,P=0.000). Conclusion The antifungal mechanism of extract of Dryopteris fragrans ( L. ) Schot may be relate to its inhibition on squalene epoxidase andβ-(1,3)-D-glucan synthase activity.
