白细胞介素-35 对乙型肝炎病毒复制的影响研究

摘要

Objective To explore the influence of interleukin-35(IL-35) on the replication of hepatitis B viru (HBV) .Methods HBV infectious clone pHBV1 .3 and its promoter pHBV-Luc were transfected into HepG2 cells respectively and different concentrations of IL-35 were added .The HBsAg and HBeAg levels in the supernatants were measured by adopting enzyme linked immunosorbent assay (ELISA) ,and the covalently closed circular DNA (cccDNA) levels were measured by real-time PCR ,and the HBV promoter activity was measured by the luminometer fluorescence detector .Results The levels of HBsAg and HBeAg ,and cccDNA copy number were inhibited by IL-35 and the HBV promoter activity was down-regulated .Conclusion IL-35 can inhibit the replication of HBV in HepG2 cells .%目的 探讨白细胞介素(IL )-35 对乙型肝炎病毒(HBV )复制的影响.方法 将 HBV感染性克隆pHBV1 .3及其启动子pHBV-Luc分别转染HepG2细胞 ,加入不同浓度的重组人IL-35蛋白 ,采用酶联免疫吸附试验(ELISA)检测细胞上清液中乙型肝炎表面抗原(HBsAg)和乙型肝炎e抗原(HBeAg)水平 ;荧光定量聚合酶链反应检测细胞闭合环状 DNA (cccDNA )的水平 ;采用 Luminometer 荧光检测仪分析 HBV 启动子活性的变化.结果 IL-35能够抑制细胞上清液中 HBsAg和HBeAg的水平及细胞内 HBV cccDNA的拷贝数 ,并下调 HBV启动子的活性.结论 IL-35能够在 HepG2细胞中抑制 HBV的复制.
