左心缺血再灌注损伤兔肺组织超微结构变化及IL-1β的表达

摘要

Aim :To study the significance of expression of IL-1 8 and changes of ultrastructure of lung tissues from left heart ischemic reperfusion rabbits. Methods: A total of 60 New Zealand rabbits were randomly divided into experimental group and control group,30 in each group. The rabbits of experimental group were copied the model of left heart ischemia reperfusion through ligating and loosing the anterior descending branch. Thirty minutes after ligating,20 minutes after loosing and 40 minutes after loosing, respectively, 10 rabbits out of each group were observed phrenic nerve discharge curve with BL-420 computer system, then were killed to observe the ultrastructure of lung tissue by transmission electron microscopy, the levels of IL-lβ in BALF and blood were detected by ELISA. Results: The phrenic nerve discharge curves of experimental group were worse than the control group( P <0.05) , the ultrastructure of lung tissue of experimental group were injuried, but these situations were not appeared in the control group. The levels of IL-1 β in BALF and blood were higher than those of the control group(P <0.05). Conclusion: IL-1β may be involved in lung injury induced by left heart ischemia reperfu-sion.%目的：探讨左心缺血再灌注损伤家兔肺组织超微结构变化及IL-1β的表达。方法：将60只成年健康家兔随机平分为实验组和对照组。实验组家兔结扎及再通左冠状动脉前降支复制左心缺血再灌注模型，对照组不结扎血管。2组均在缺血30 min、再灌注20 min、再灌注40min 3个时间点取10只动物,描记膈神经放电曲线；取外周血和支气管肺泡灌洗液(BALF)，ELISA法检测外周血和BALF中的IL-1β水平；取肺组织标本，电镜下观察肺组织超微结构。结果：与对照组比较，实验组各时间点膈神经放电幅度减小(P＜0.001)，放电持续时间缩短(P＜0.001)；电镜下实验组可见Ⅰ型和Ⅱ型肺泡上皮细胞变性，呈急性炎症损伤改变，对照组无类似改变；实验组外周血和BALF中IL-1β表达水平均高于对照组(P＜0.05)。结论：IL-1β参与了左心缺血再灌注所致的肺损伤。
