侵袭性牙周炎患者牙龈组织中MMP-8及TIMP-1的表达

摘要

目的:探讨基质金属蛋白酶-8(MMP-8)及其抑制因子-1 (TIMP-1)在侵袭性牙周炎(AgP)牙龈组织中的表达情况.方法:采用免疫组织化学方法检测15例AgP患者和20例对照的牙龈组织中MMP-8和TIMP-1蛋白的表达情况.结果:MMP-8和TIMP-1在2组牙龈组织的上皮内及上皮下结缔组织中均有表达.AgP组MMP-8表达水平和MMP-8/TIMP-1均高于对照组(Z=2.990和4.911,P<0.05),2组TIMP-1的表达差异无统计学意义(Z=1.822,P>0.05).ROC曲线分析结果显示,AUC(MMP-8)=0.752(95%CI=0.618~0.885,P=0.003),AUC(MMP-8/TIMP-1)=0.913(95%CI=0.831~0.996,P<0.001),AUC(TIMP-1)=0.347(95%CI=0.194~0.499,P=0.068).结论:牙龈组织中MMP-8表达水平和MMP-8/TIMP-1可以作为诊断AgP的潜在指标.%Aim:To study the expression of matrix metalloproteinase -8( MMP-8) and its inhibitory factor4 ( TIMP4 ) in gingival tissue of patients with aggressive periodontitis (AgP).Methods: Immunohistochemistry method was used to detect MMP-8 and TIMP4 protein in gingival tissue of 15 AgP patients and 20 healthy control.Results : MMP-8 and TIMP4 were positively expressed in the intraepithelial and subcutaneous connective tissue in both groups .MMP-8 expression level and MMP-8/TIMP4 of AgP group were significantly higher than those of the control group ( Z = 2.990 and 4.911, P < 0.05 ) , but the difference in TIMP4 expression between the 2 groups was not significant ( Z = 1.822 , P > 0.05 ).ROC curve analysis showed that AUC( MMP-8) = 0.752 (95% CI = 0.618-0.885, P = 0.003), AUC ( MMP-8/TIMP4) = 0.913 (95% CI = 0.831-0.996 ,P < 0.001) ,AUC(TMP-1) = 0.347(95% CI = 0.194 - 0.499,P= 0.068).Conclusion: MMP-8 expression level and MMP -8/TIMP4 in gingival tissue could be helpful for the diagnosis of AgP .
