目的:观察在牙齿移动过程中三七总皂苷(PNS)对兔牙周组织转化生长因子-β1(TGF-β1)表达的影响.方法:选用3~6月龄的雄性新西兰兔16只,随机分成PNS组和生理盐水(NS)组,通过拉簧分别加力于下颌双侧第一前臼齿与门牙之间,建立兔牙齿移动模型,然后两组分别肌内注射血栓通[10 mg/(kg·d)]和NS.建模前1周,建模后1、2、3、4周后分别采血,全自动生化分析仪检测血清中钙离子、磷离子以及碱性磷酸酶的含量.建模后第4周处死兔,取移动牙齿及其牙周组织,采用免疫组化染色和免疫荧光染色方法检测TGF-β1的表达.结果:PNS组血清中碱性磷酸酶的含量高于NS组(P<0.05),牙周组织中TGF-β1的表达高于NS组(P<0.05).结论:在牙齿移动过程中,PNS能增加兔牙周组织中TGF-β1的表达,对牙槽骨的改建有一定的促进作用.%Aim:To investigate the effect of panax notoginseng saponins(PNS) on the expression of transforming growth factor-β1(TGF-β1) in periodontal tissue of rabbits during teeth movement.Methods: Sixteen 3-6-month-old male New Zealand rabbits were randomly allocated into two groups.The rabbits of PNS group were injected Xueshuantong and the rabbits of normal saline group were injected normal saline.Two tension springs with 80 g force were respectively applied between the first premolars and mandibular anterior teeth.After four weeks raising,the rabbits were sacrificed for the first premolars and periodontal tissues.Immunohistochemical staining and immunofluorescence staining were used to detect the expression of TGF-β1 in periodontal tissues of the teeth.At the day before modeling,and the 1st,2nd,3rd,and 4th week after modeling,blood samples were collected.The concentration of calciumion,phosphorus ion and alkaline phosphatase in serum was determined with automatic biochemical analyzer.Results: The concentration of alkaline phosphatase in serum in PNS group was higher than that in normal saline group(P<0.05).The expression of TGF-β1 of PNS group was stronger than normal saline group(P<0.05).Conclusion: Panax notoginseng saponins can promote the expression of TGF-β1 in the periodontal tissue of rabbits,and may play a certain role in promoting alveolar bone remodeling during teeth movement.
