三种南方贝母的RAPD分析

         

摘要

利用RAPD技术对三种南方贝母品种浙贝母、川贝母和湖北贝母进行分析.RAPD扩增结果显示扩增产物的分子量介于100-2300 bp之间,共扩增出39条谱带,多态性条带数在6-10条范围内.引物S303的扩增效果最好,稳定性亦较高,其扩增产物的多态性分布图可以作为浙贝母的中药分子鉴定依据.利用RAPD分析三种南方贝母的遗传关系,发现浙贝母与川贝母亲缘关系较近,和湖北贝母亲缘关系较远,其亲缘关系的远近与地理分布相似.%RAPD was used to study genetic polymorphism and relationships among F. Cirrhosa, F. Thunbergii Miq. And F. Hupehensis Hsiao et in southern China. The results showed that 39 bands were clearly displayed with molecular weights ranging fromlOO bp to 2 300 bp. Every primer could amplify 6 to 10 polymorphic bands. Primer S303 showed better polymorphism and stability. Its amplification pattern could be used to identify F. Thunbergii Miq. We also used RAPD to study the genetic relationships among three species of Fritillaria in southern China. The F. Thunbergii Miq. And F. Cirrhosa had the closest relationship to each other, while F. Thunbergii Miq. And F. Hupehensis Hsiao et had the farthest one. Their hereditary relationship is noted by the geographical distribution.
