橙皮苷对脂肪乳剂致高脂血症模型大鼠血脂及血液流变学的影响

         

摘要

[目的]探讨橙皮苷对脂肪乳剂灌胃致高脂血症模型大鼠的血脂及血液流变学的影响.[方法]采用脂肪乳剂连续灌胃40d致SD大鼠高脂血症模型,连续灌胃给予橙皮苷40d.给药第2、4周各眼眶取血一次,测定大鼠血清总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-c)、低密度脂蛋白胆固醇(LDL-c)含量并计算动脉粥样硬化指数(AI);末次给药后下腔静脉取血,测定血液流变学指标.[结果]与模型对照组比较,给药2周及4周后,橙皮苷三个剂量(60mg·kg-1、120mg·kg-1、240mg·kg-1)组大鼠血清TC、LDL-c、AI明显降低,120 mg·kg-1、240mg· kg-1剂量组大鼠HDL-c升高;给药40d后,橙皮苷三个剂量(60 mg·kg-1、120 mg·kg-1、240mg·kg-1)组高脂血症大鼠各切变率下全血黏度、血浆黏度、红细胞压积、红细胞计数、血沉方程K值、卡松屈服应力及红细胞电泳时间明显降低,红细胞变形指数明显升高,且120mg· kg-1、240mg· kg-1剂量作用明显.[结论]橙皮苷60mg· kg-1、120mg· kg-1、240mg· kg-1三个剂量均可调节高脂血症模型大鼠血脂水平及血液粘滞状态,且120mg· kg-1、240mg· kg-1剂量作用明显,提示橙皮苷对防治高脂血症具有积极作用.%[Objective]To research the effect of Hesperidin on blood fat and hemorheology in hyperlipidemia rat by fat emulsion. [Methods ]Sprague Daw-ley rats were administered with fat emulsion for 40 days consecutively to establish hyperlipidemia model. The treatment groups were simultaneously administered with Hesperidin for 40 days. The blood was collected to separate into the serum, in which the levels of cholesterol(TC), high density lipopro-tein-cholesterol(HDL-c),low density lipoprotein-cholesterol(LDL-c) were measured and atherosclerosis in dex(AI) and were calculated at the 2th and 4th week. After the last administration.the blood was taken from inferior vena cava, in which the levels of hemorheology were detected. [Results]Compared with model control, the level of the serum TC,LDL-c,Al all decreased at the 60mg·kg-1,120mg·kg-1,240mg·kg-1 dose groups. So it was after 4 weeks. Hesperidin at the 60 mg·kg-1 120 mg·kg-1 and 240 mg·kg-1 dose groups could markedly reduce all kinds of speed-whole blood viscosity, plasma viscosity, hematokrit, red blood cell count, sedimentation equation K value, KaSong yield stress and erythrocyte electrophoresis time, especially at the 120mg·kg-1 X 240mg· kg-1 dose groups. [Conclusion]The blood fat and hemorheology in hyperlipidemia rat could be regulated by three doses of Hesperidin,what is more.the results were better at 120 mg·kg-1 dose and 240mg·kg-1 doses group. It was indicated that hesperidin had a positive role in the prevention and treatment of hyperlipidemia.
