Tim-3/Galectin-9与 Th1/Th2相关因子在人细粒棘球蚴感染中的表达研究

     

摘要

目的:探讨人细粒棘球蚴感染中 Tim-3/Galectin-9与 Thl/Th2的平衡关系及其与该疾病发生、发展的关系。方法用实时荧光定量 PCR 法分别检测30例慢性细粒棘球蚴感染者和30例健康体检人群外周血中Tim-3、Galectin-9、T-bet 和 GATA-3 mRNA 的表达,同时用酶联免疫吸附(ELISA)法检测慢性细粒棘球蚴感染者和健康对照者血清中可溶性 Tim-3和 Galectin-9及 IFN-γ、IL-12和 IL-4的水平。结果(1)实时荧光定量 PCR检测结果显示:与健康对照组相比,感染组 Tim-3和 Galectin-9 mRNA 水平显著升高,T-bet mRNA 水平显著降低,GATA-3 mRNA 水平显著升高。(2)ELISA 检测结果显示:与健康对照组相比,感染组血清中可溶性 Tim-3和Galectin-9水平显著升高,IFN-γ和 IL-12水平显著下降,IL-4水平显著升高。Tim-3与 Galectin-9呈正相关(P =0.000,r =0.776);Tim-3与 T-bet 呈负相关(P =0.000,r =-0.645);Tim-3与 GATA-3呈正相关(P =0.000,r=-0.752)。结论 Tim-3/Galectin-9在人细粒棘球蚴持续性感染中发挥一定的作用,Tim-3/Galectin-9可能通过影响 Th1/Th2相关的转录因子 T-bet/GATA-3和细胞因子 IL-12、IFN-γ和 IL-4的水平变化,促进 Th1/Th2细胞失衡现象的发生,从而促进感染的持续性发展。%Objective To explore role of relationship between Tim-3/Galectin-9 and Thl/Th2 balances in Echinococcus granululosus infected development by studying relationship between Tim-3,Galectin-9 and Th1/Th2 related transcription factors T-bet/GATA-3 and related cytokines IFN-gamma,IL-12,IL-4 in peripheral blood.Methods Utilize the real-time quantitative PCR technique to detect Tim-3,Galectin-9, T-bet and GATA-3 mRNA expression in 30 cases of chronic Echinococcus granululosus infection patients vs 30 cases of healthy control.At the same time,use enzyme linked immunosorbent assay (ELISA)meth-od to detect the levels of serum soluble Galectin-9,Tim-3,IFN-gamma,IL-12 and IL-4 in chronic Echinococcus granululosus infection patients and healthy control respectively.Results (1)RT-PCR detec-tion showed that compared with healthy control,the mRNA levels of Tim-3 and Galectin-9 in chronic in-fection group were increased significantly;The mRNA level of T-bet is decreased significantly;The mRNA level of GATA-3 is increase significantly.(2)ELISA test showed that compared with healthy con-trol,the chronic infection group serum soluble Galectin-9 and Tim-3 levels increased significantly;IFN-gamma and IL-12 levels decreased significantly;IL-4 levels increased significantly.(3)Tim-3 was positive-ly correlated with Galectin-9 (P = 0.000,r =0.776);Tim-3 and T-bet were negatively correlated (P = 0.000,r=-0.645);Tim-3 and GATA-3 were positively correlated (P = 0.000,r =-0.752).Conclusion Tim-3/ Ga-lectin-9 play an important role in Echinococcus granululosus persistent infection,and Tim-3/Galectin-9 may pro-mote the imbalance of Th1/Th2 cells by affecting Th1/Th2 related transcription factors T-bet/GATA-3 and cyto-kine IL-12,IFN-gamma and IL-4 levels to promote the sustainable development of the infection.
