散癖平胃方对人胃癌细胞SGC-7901侵袭转移的影响

         

摘要

目的 观察散癖平胃(SPPW)方对人胃癌细胞SGC-7901侵袭转移能力的影响.方法 SPPW方含药血清干预人胃癌细胞SGC-7901 48h后,应用黏附实验、侵袭实验及迁移实验,检测SPPW方对人胃癌细胞SGC-7901侵袭转移能力的影响.应用实时定量聚合酶链反应( real-time PCR)检测SPPW对与侵袭转移有关的MMP-2和MMP-9mRNA表达的影响.结果 SPPW方各组含药血清干预人胃癌细胞48 h后,SGC-7901细胞黏附抑制率、侵袭抑制率及迁移抑制率均增加且呈剂量依赖关系,与阴性对照组比较有统计学差异(P<0.01或P<0.05).SPPW方三个剂量组对MMP-2 mRNA的表达无统计学差异,对MMP-9 mRNA的表达具有统计学差异(P<0.01).结论 SPPW方可抑制人胃癌细胞SGC-7901黏附、侵袭和迁移能力,其抑制有明显的剂量依赖性,其抑制的机制可能与下调MMP-9的表达有关.%Objective To investigate the effects of Sanpipingwei Formula (SPPW) on invasion and metastasis of gastric cancer SGC-7901 cells in vitro. Methods After 48-hour treatment with serum containing traditional Chinese medicine SPPW, the effect on adhesion of SGC-7901 cells was tested by cell matrigel adhesion assay. Transwell chamber assay was performed to determine the effect on invasion and migratory capacity of the cells. The level of MMP-2 and MMP-9 mRNA in the cells was measured with real-time PCR. Results After 48-hour treatment with serum containing SPPW, the inhibition rates of adhesion, invasion and migration of SGC-7901 cells were increased in a dose-dependent manner and significantly differed from those in negative control group (P<0.01 or P<0. 05). Compared with that in negative control group, MMP-2 mRNA level in SPPW groups had no significant difference. However, MMP-9 mRNA level was restrained in SPPW groups (P<0.01). Conclusion The abilities of adhesion, invasion and metastasis of SGC-7901 cells are visibly inhibited by SPPW in a dose-dependent manner. SPPW's inhibitory effect may be related to its down-regulating the MMP-9 mRNA expression.
