硫化氢对大鼠离体结肠平滑肌细胞作用的研究

         

摘要

Objective: To study the effect of hydrogen sulfide (H2S) on the size of single colonic smooth muscle cell in vitro and the molecule mechanism. Methods: Smooth muscle cells were isolated from the colon of rat.The relaxation response of colonic smooth muscle cells to sodium hydrosuliide (NaSH)(H2S donor) and other medicines were separately observed. Results:(1)The relaxation value of cells treated with NaSH at lx10-3 and lx10-4 mol/L were 11.49%and 20.57%.(2)The relaxation induced by NaSH could be attenuated by glibenclamide (Gli) ( KATP channel blocker ). But the relaxation effect of NaSH could not be blocked by Gli completely. (3)Pinacidil (Pin) (KATP channel opener) had the same effect on cells relaxation. Conclusion: H2S may produce the relaxation effect on colonic smooth muscle cells, which maybe partly due to the activation of KATP channel.%目的:研究硫化氢( H2S)对结肠平滑肌细胞舒缩作用的影响及其作用机制.方法:制备大鼠离体的结肠平滑肌细胞,观察NaSH(H2S供体)的舒张效应及其他工具药对该舒张效应的影响.结果:(1)1×10-5 mol/L、1×10-4 mol/L NaSH分别可使结肠平滑肌细胞舒张11.49%、20.57%.(2)ATP敏感钾离子通道(KATP通道)阻滞剂格列本脲能减弱NaSH的细胞舒张作用,但不能完全阻断.(3)KATP通道开放剂吡那地尔也可产生细胞舒张作用.结论:H2S可以使离体单个结肠平滑肌细胞产生舒张作用;激活KATP通道可能是其作用机制之一.
