U73122和SQ22536对豚鼠视网膜色素上皮细胞分泌bFGF的影响

Objective: To evaluate the roles of phospholipase C inhibitor U73122 and adenylyl cyclase inhibitor SQ22536 on the function of secreting bFGF in the cultured guinea pig's retinal pigment epithelial (RPE) cells.Methods: RPE cells were from the guinea pig eyes,the second generation was used to the experiment.The drug U73122 or SQ22536 with the concentration of l×10-5mol／L was added to the culture solution in the experimental group.And the control groups were added the same dose of DMSO or ddH20.At the times of 2,4,6,8,16,24,48hours ,collected the culture solution and the bFGF secreted by RPE cells was tested using an ELISA method.Results: In the U73122 group, the result of the ELISA test showed that the secretion of bFGF was decreased at the times of 2,4,6,8,16 hours, compared with the control groups (P＜0.05).But,at the times of 24,48 hours, the secretion of bFGF was no statistical significance between the drug group and the control group (P＞0.05).In the SQ22536 group, the ELISA test showed that the secretion of bFGF was decreased at the limes of 2,4,6,8,16,24,48 hours compared with the control group(P＜0.05).Conclusion: The phospholipase C inhibitor, U73122 and the adenylyl cyclase inhibitor SQ22536 can inhibit the secretion of bFGF in RPE cells.It shows that the secretion of bFGF may be regulated by the adenylyl cyclase sigua transduction system and the phospholipase C signal transduction system in the RPE cells.%目的:观察磷脂酶C抑制剂U73122和腺苷酸环化酶抑制剂SQ22536对豚鼠视网膜色素上皮(RPE)细胞分泌碱性成纤维细胞生长因子(bFGF)的影响.方法:培养原代豚鼠RPE细胞,传2代后用于实验.用含U73122及SQ22536浓度均为l×10-5mol/L的两种培养液培养RPE细胞,设含有单纯溶质的培养液作为对照.分别于实验2、4、6、8、16、24、48h收集培养液,ELISA法检测bFGF的含量.结果:ELISA结果显示:加入1×10-5mol/ L U73122后,实验组bFGF的分泌量与对照组相比在2、4、6、8、16h减少(P＜0.05),24、48h变化无统计意义(P＞0.05);而SQ22536组与对照组相比RPE细胞bFGF的分泌量在各时间点均有减少(P＜0.05).结论:U73122和SQ22536对RPE细胞bFGF的分泌均有抑制作用.提示RPE细胞内可能是通过磷脂酶C信号途径和腺苷酸环化酶信号途径共同调控bFGF的分泌.
