基于响应面分析法的超声波洗涤羽毛纤维工艺条件优化

             

摘要

通过洗涤降低羽毛纤维上含菌量是其前处理的重要任务.以洗涤时间、洗涤温度、浴比和酸性蛋白酶用量为自变量,洗净羽毛纤维上嗜温性需氧菌含量为响应值,采用响应面分析法对羽毛纤维的洗涤工艺条件进行优化.通过单因素试验、Box-Benhnken中心组合试验和响应面分析法研究各自变量及其交互作用对含菌量的影响,得到二次多项式回归方程的预测模型,确定超声波洗涤羽毛纤维的最佳工艺条件为:双氧水浓度5 mL/L、pH值4.0~6.0、洗涤时间20 min、洗涤温度60℃、浴比1∶40、酸性蛋白酶用量3%(o.w.f).按此最优工艺条件洗涤后,羽毛纤维上嗜温性需氧菌的含量为3.73×103 cfu/g.%It is the important task in pretreatment to reduce bacterial content of feather fibers by washing. Process optimization of ultrasonic washing of feather fibers was performed by response surface method in which washing time, washing temperature, liquor ratio and dosage of acid protease were used as independent variables and the mesophilic aerobic bacterial content of feather fibers was used as the response surface. The effects of each independent variable and their interaction on the mesophilic aerobic bacterial content were investigated using single factor analysis, Box-Benhnken center composite design and response surface method, and the mathematical regression equation prediction model was established. The optimum washing conditions are as follows; hydrogen peroxide solution 5 mL/L, pH value 4. 0 -6.0, washing time 20 min, washing temperature 60 ℃ , liquor ratio 1: 40 and dosage of acid protease 3% ( o. w. f) . Under these conditions, the content of mesophilic aerobic bacteria of washed feather fibers is 3. 73 × 103 cfu/g.
