离子导入巩膜交联术对近视模型力学性能及胶原代谢的影响

             

摘要

Scleral collagen cross-linking (SXL)is one of the most promising treatments to control the pathologic process of myopia.The exact procedure and effects on myopic animal mod-els still need to be explored.We applied an iontophoresis-assisted delivery system to the ribofla-vin/ultraviolet A (UVA)SXL procedure,and conducted an accelerated UVA irradiation (10 mW/cm2 ,9 minutes)on a myopic animal model to explore the effects on scleral biomechanical properties and collagen metabolism.By form deprivation,ninety-six New Zealand white rabbits developed relatively stable myopia and then underwent the SXL surgery or sham-operation.All the sample and statistics were obtained from 4 time points (1-day,10-day,1-month and 3-month) after the surgery.The results show that the ultimate stress,Young's modulus and physiological Young's modulus of treated myopic sclera were significantly increased compared with those of un-treated ones in all four groups.In addition,there were significant differences in the expression of genes and proteins related to collagen metabolism between the treated sclera samples and untreat-ed ones.It demonstrated that iontophoresis-assisted accelerated Riboflavin/UVA SXL might be a potential method to control the pathological process of myopia.%巩膜胶原交联术是有望控制近视病程发展的方法之一.然而,该术式的确切程序及其对近视动物模型的影响仍有待探讨.将离子导入法引入巩膜核黄素-紫外线A(UVA)交联术,采用10 mW/cm2,9 min的快速交联方案,研究该术式对于近视动物模型巩膜生物力学性能及胶原代谢的影响.利用形觉剥夺法将96只新西兰大白兔诱导形成相对稳定的近视模型,然后进行巩膜交联术或假手术,术后4个时间点(1 d,10 d,1个月,3个月)分别采集样本和获取数据.术后4个时间点,与非交联近视眼相比,交联近视眼巩膜的极限应力、杨氏模量和生理杨氏模量均明显增加.此外,交联组与未交联组胶原代谢相关基因和蛋白的表达存在显著的差异.结果表明,离子导入核黄素-紫外线A快速交联术很可能是一种控制近视病理过程的可行性方法.
