RP-HPLC法测定西红花甙提取物中西红花甙-1和西红花甙-2的含量

         

摘要

Objective To apply RP-HPLC method to determine the content of crocusⅠ and crocusⅡ in crocus sativus L.Methods The HPLC instrument was equipped with Nova-Pak C18(4μm,150mm×3.9mm) column.The mobile phase was methanol-water(55∶45).The flow rate was 1ml*min-1 .The UV detector wavelength was set at 440nm.Results The standard curve of crocusⅠwas linear in the concentration range of 0.03~0.30g*L-1(r=0.9999),the average recovery rate was 100.21% and relative standard was 0.38%(n=5).The standard curve of crocus Ⅱ was linear in the concentration range of 0.05~0.30 g*L-1(r=0.9999),the average recovery rate was 99.78% and relative standard was 0.42% (n=5).Conclusion The method is simple,sensitive,and accurate to determine the content of crocusⅠand crocus Ⅱ in crocus L.%目的:建立反相高效液相色谱测定西红花甙提取物中西红花甙-1和西红花甙-2含量的方法。方法:采用Nova-Pak C18(4μm,150mm×3.9mm) 色谱柱,流动相甲醇-水(55∶45),流速1ml*min-1,检测波长440nm。结果:西红花甙-1对照品的线性范围为0.03~0.30g*L-1,r=0.9999,平均加样回收率为100.21%,RSD=0.38%;西红花甙-2对照品的线性范围为0.05~0.30g*L-1,r=0.9999,平均加样回收率为99.78%,RSD=0.42%。结论:采用反相高效液相色谱法测定西红花提取物中西红花甙-1和西红花甙-2含量,操作简便、结果准确。
