神经干细胞移植对脑缺血再灌注损伤大鼠脑脊液中炎症因子的影响

         

摘要

Objective: To investigate the effects of neural stem cells (NSCs) on inflammatory response in rats with cerebral ischemia-reperfusion model.Methods: Ischemia-reperfusion model of rats was built by middle cerebral ar-tery occlusion and reperfusion (MCAO/R, occluded 90 min) method.Rats were randomly divided into sham opera-tion group, phosphate buffer(PBS) control group, and NSCs transplantation group.Then on the 1st, 3rd, 14th and 28th days after stroke and transplantation , the neurological functions were assessed by m-NSS score, and the cere-brospinal fluid was collected and then detected by ELISA.Results: (1) The neurological function of NSCs trans-plantation group was significantly improved than that of PBS group with statistically significant difference ( P <0.05).(2)The levels of TNF-αand IL-1βin cerebrospinal fluid of NSCs transplantation group were lower than those in the control group on the 1st, 3rd and 14th days, and the levels of TGF-β1 and IL-10 in NSCs group were higher than those in the control group , the difference being statistically significant (P<0.05).Conclusion: Trans-plantation of NSCs can alleviate the inflammatory response after cerebral ischemia-reperfusion injury in rats.%目的:探索神经干细胞( NSCs)移植对大鼠脑缺血再灌注损伤后炎症反应的影响.方法:SD大鼠随机分为假手术组、磷酸盐缓冲液(PBS)对照组、NSCs移植实验组,对照组和实验组构建大脑中动脉闭塞再灌注(MCAO/R)模型,然后分别立体定位注射PBS和NSCs,在1、3、14、28d对各组动物进行m-NSS评分和取脑脊液行ELISA检测.结果:(1)对照组、实验组大鼠神经功能都有恢复,而实验组更显著,m-NSS评分差异具有统计学意义(P<0.05). (2)实验组脑脊液中肿瘤坏死因子α、白细胞介素1β含量在1、3、14d都低于对照组,相对应的转化生长因子β1、白细胞介素10含量均高于对照组,差异均具有统计学意义( P<0.05).结论:NSCs移植能抑制大鼠脑缺血再灌注损伤后炎症反应.
