濒危药用植物天山雪莲的根段组织培养与植株再生

     

摘要

An efficient micro-propagation system for Saussurea involucrata Kar. Et Kir. An endangered Chinese medicinal plant, has been developed. 100% callus formation frequency was got from root explants inoculated on a MS medium containing 0. 5 mg/L 2,4-D and 2 mg/L NAA (1-Naphthaleneacetic acid). 42. 6% of adventitious bud organogenesis frequency and 4. 3 buds per explant were got from root-derived callus when cultured on a MS medium supplemented with 0. 05 mg/L NAA.0. 4 mg/L 6-BA. Up to 76. 9% of the regenerated shoots formed complete roots on a half-strength MS medium containing 0. 3 mg/L NAA,and 89% of the complete plantlets survived and grew vigorously under greenhouse condition.%以濒危药用植物天山雪莲根段为研究材料进行组织培养和植株再生研究,旨在拓宽天山雪莲组织培养外植体的范围,建立高效植株再生体系,为其种群复壮、资源保护与可持续利用提供技术参考.研究结果表明:适合天山雪莲根段愈伤组织诱导的最佳培养基为MS+2,4-D(0.5 mg/L) +NAA(2.0 mg/L),愈伤组织的诱导率为1000%;适合其根段愈伤组织从生芽分化的最佳培养基为MS+ NAA(0.05 mg/L)+6-BA(0.4 mg/L),分化率为42.60,增殖倍数为4.3;最佳生根培养基为1/2 MS+NAA(0.3 mg/L),在此条件下,根发育良好,生根率为76.9%,植株健壮;组培苗炼苗后移栽,成活率可达89%.
