不同培养条件下蜡蚧轮枝菌VL17分生孢子的耐热力及孢壁疏水蛋白含量的分析

         

摘要

比较了不同培养条件下蜡蚧轮枝菌VL17分生孢子耐热力及其孢壁疏水蛋白(FAE)含量的变化,结果表明:葡萄糖为碳源,酵母浸膏为氮源时孢子表现出较强的耐热力,残存指数(Is)分别为0.72和0.69;而麦芽糖为碳源,酵母浸膏为氮源时,FAE蛋白含量高达28.7μg· mg-1和24.7μg·mg-1;培养基中添加50μg· mL-1Mn2+时所产孢子的耐热力最强,Is为0.68,而Zn2+浓度50μg.mL-1的培养基所产孢子FAE蛋白的含量最高,为19.9μg· mg-1;培养基pH值在6~7时所产孢子的耐热力及FEA蛋白含量都处于较高水平,Is值为0.51,FAE含量为24.3μg· mg-1;30℃培养条件下所获孢子的耐热力显著高于常温所产孢子,Is值为0.57,但FAE蛋白含量较常温所产孢子略有下降.%Conidial terotolerance of Vertkillium lecanill VL17 and concentration of FAE proteins were investigated in different medium and culture conditions. The results showed that glucose was optimal carbon sources for improving conidial heathardiness and the best nirtrogen sources is yeast extract. They survival index were 0.72 and 0.69. When maltose as carbon source and yeast extract as nitrogen source, concentration of FAE proteins achieved a higher level (28.7μg-mg-1 and 24.7μg·ng-1). Conidial terotolerance reached a higher level which came from the medium containing 50ug·mL-1 Mn2+, survival index was 0.68. When the medium contained 50(gTnL-1 Zn2+ concentration of FAE proteins was as high as 19.99μg·mg-1. The medium with pH between 6 and 7 could enhance conidial therotolerance and increase FAE proteins content, survival index was 0.51, concentration of FAE was 24.3u.g-mg-1 These conidial which produced at 30t had a higher heat resistant, survival index was 0.37, but the content of FAE proteins was lower than that produced at the room temperature.
