目的 建立地锦草总黄酮含量的测定方法及优化地锦草中总黄酮的提取工艺.方法 采用紫外-可见分光光度法测定总黄酮含量;提取工艺考察了水回流提取、乙醇回流提取及超声提取不同的提取方法,优选出乙醇回流提取法,采用单因素试验及正交试验优化地锦草的乙醇回流提取工艺.结果 该测定方法的平均回收率为102.82%,RSD=1.2%;地锦草总黄酮提取工艺考察,最终确定最佳工艺:75%乙醇,15倍量,提取2次,每次2 h;提取率高达到2.588%.结论 优选的总黄酮提取工艺方法简单、易行.%Objective To establish a ultraviolet-visible spectrophotometry for determining the content of total flavonoids in Euphorbia humifusae and to optimize its extraction conditions. Methods Ultraviolet-visible spectrophotometry was used to determine the content of total flavonoids. The optimum extraction conditions were explored by single factor analysis and orthogonal design with the water extraction, alcohol extraction and ultrasonic extraction method as factors. Results The average recovery rate was 102. 82% with RSD 1.2%. The optimum conditions were 75% ethanol, ratio of ethanol to material of 15, extraction for 2 times and 2 h each time. The extraction rate of total flavonoids under the optimum conditions was 2. 588% . Conclusion The established method was simple and feasible for gaining the high extraction rate.
