Wnt信号通路中GSK-3β和CD44 v6在人结直肠肿瘤组织中的表达及意义

         

摘要

目的:探讨Wnt信号通路中GSK-3β、CD44v6在人结直肠肿瘤组织中的表达及其相关性。方法应用免疫组化Envision法测定GSK-3β和CD44v6在正常结直肠黏膜组(n=20),结直肠腺瘤组(n=19)和腺癌组(n=45)中的表达情况,分析GSK-3β和CD44v6在结直肠癌中的表达及与临床病理参数之间的关系及其相关性。结果①GSK-3β在正常肠黏膜、结直肠腺瘤及结直肠癌三组中均表达,阳性表达率分别为5.0%(1/20),42.1%(8/19),77.8%(35/45)。癌组织中GSK-3β表达显著高于腺瘤组和正常黏膜组(P <0.05),GSK-3β表达与 Dukes 分期、分化程度、有无淋巴结转移均无关(P >0.05)。②CD44v6在癌组织中阳性表达率55.6%(25/45),在结直肠腺瘤及正常肠黏膜中阳性率为0,在癌组织中表达显著高于结直肠腺瘤组和正常黏膜组(P<0.05)。 CD44v6的表达与 Dukes分期、淋巴结转移相关(P <0.05),而与分化程度无关。③经Spearman秩相关分析,GSK-3β与CD44v6在结直肠癌中的表达无相关性(r=0.167,P>0.05)。结论 GSK-3β和CD44v6均参与了结直肠肿瘤的发生及发展,未来可能是判断肿瘤预后的标志。 CD44v6在结直肠癌的浸润转移过程中可能起着重要的作用。%Objective To explore the GSK-3βand CD44v6 expression of Wnt signaling pathway in human colorectal tumors and their relationship. Methods The expression of GSK-3β and CD44v6 proteins was detected in 20 cases of normal colorectal mucosa,19 cases of colorectal adenomas and 40 cases of colorectal carcinoma using EnVision immunohistochemistry. The relationships of GSK-3βand CD44v6 with the clinicopathological parameters were analyzed in colorectal adenomas. Results ①The positive rates of GSK-3βin normal intestinal mucosa, colorectal adenoma and colorectal carcinoma were 5. 0%(1/20),42. 1%(8/19) and 77. 8%(35/45), re-spectively. The expression of GSK-3βin colorectal carcinoma was significantly higher than those in the colorectal adenoma and normal intestinal mucosa(P<0. 05). The expression of the GSK-3βin colorectal carcinoma was not correlated with Dukes stage, histological differentiation and lymph node metastasis(P>0. 05). ②The positive expression of CD44v6 was found in 25 cases(55. 6%) in the colorectal adenomas, but not found in the colorectal adenoma and normal intestinal mucosa. The expression of CD44v6 in colorectal carcinoma was significantly higher than that in the colorectal adenoma and normal intestinal mucosa(P<0. 05). The expression of CD44v6 in colorectal carcinoma was correlated with Dukes stage and lymph node metastasis(P<0. 05), but not correlated with histo-logical differentiation(P>0. 05). ③Spearman correlation analysis showed that the expression of GSK-3β was not correlated with CD44v6 expression(r=0. 167,P>0. 05). Conclusion The GSK-3βand CD44v6 are involved in the occurrence and development of colorectal tumor, and may be markers of tumor prognosis in future. CD44v6 may play an important role in the infiltration and metastasis of colorectal carcinoma.
