氯胺酮联用硫酸镁对小鼠学习记忆的影响

         

摘要

Objective To investigate the effect of ketamine combined with magnesium sulfate on learning and memory in mice.Methods Eighty KM mice aged six weeks were divided into four groups (n =20 in each group):ketamine group,magnesium sulfate group,ketamine and magnesium sulfate group,normal saline control group(NS group).The mice were treated by intraperitoneal injection of normal saline(0.1 ml/10 g) or magnesium sulfate(15 mg/kg) and subcutaneous injection of ketamine(50 mg/kg),respectively.The error times and step down latency(SDL) in step down test,and error times and step through latency(STL) in step through test were observed and recorded.Results In step down test,the error times and SDL showed no statistically significant difference at the day of injection between ketamine and magnesium sulfate group and ketamine group [1.6 ± 1.0 vs 1.5 ± 0.5,P > 0.05;(70 ± 40) s vs (72 ± 43)s,P > 0.05].The error times in ketamine group was more than that of magnesium sulfate group (1.5 ± 0.5 vs 0.8 ± 0.6,P < 0.05).SDL in ketamine group was lower than that in magnesium sulfate group[(72 ± 43) s vs (120 ± 50) s,P < 0.05].At the second day after injection,the error times and SDL of all mice had no difference (P > 0.05).In step through test,the error times and STL in ketamine and magnesium sulfate group at the day of injection showed no statistically significant difference compared with ketamine group [2.8 ± 1.0 vs 2.6 ± 1.4,P > 0.05;(105 ± 102) s vs (110 ± 80) s,P > 0.05].The error times in ketamine group was more than that in magnesium sulfate group (2.6 ± 1.4 vs 1.2 ± 1.5,P < 0.05).STL in ketamine group was lower than that in magnesium sulfate group [(110 ±80)s vs (214 ± 107)s,P <0.05].At the second day after injection,the error times and STL of all mice showed no difference (P > 0.05).Conclusion Ketamine can depress the learning and memory at the early stage,but ketamine and magnesium sulfate has no synergic inhibition effect on learning and memory in mice.%目的 观察氯胺酮(ketamine)与硫酸镁(magnesium sulfate)合用对小鼠学习记忆的影响.方法 6周龄昆明小鼠80只,随机分为氯胺酮组、硫酸镁组、氯胺酮和硫酸镁联合用药组和生理盐水对照组(NS组),每组20只,雌雄各半.生理盐水注射量为0.1 ml/10 g,硫酸镁腹腔注射量为15 mg/kg,氯胺酮皮下注射量为50 mg/kg.采用跳台实验和避暗实验观察药物对小鼠跳台潜伏期(step down latency,SDL)、步入潜伏期(step through latency,STL)和错误次数(error times)的影响.结果 跳台实验用药第1天,联合用药组错误次数和SDL与氯胺酮组比较,差异无统计学意义[1.6±1.0 vs 1.5±0.5,P>0.05;(70±40)s vs (72±43)s,P>0.05];氯胺酮组错误次数多于硫酸镁组(1.5±0.5 vs0.8±0.6,P<0.05),氯胺酮组SDL短于硫酸镁组[(72±43)s vs (120±50)s,P<0.05];用药第2天四组大鼠跳台实验错误次数和潜伏期差异无统计学意义(P>0.05).避暗实验用药第1天联合用药组错误次数和STL与氯胺酮组相比差异无统计学意义[2.8±1.0 vs 2.6±1.4,P>0.05;(105±102)s vs (110±80)s,P>0.05];氯胺酮组错误次数多于硫酸镁组(2.6±1.4 vs 1.2±1.5,P<0.05),氯胺酮组潜伏期短于硫酸镁组[(110±80)s vs (214±107)s,P<0.05];用药第2天四组大鼠避暗实验错误次数和潜伏期差异无统计学意义(P>0.05).结论 氯胺酮注射初期可抑制小鼠学习记忆,而硫酸镁与氯胺酮合用对学习记忆的影响无协同作用.
