Objective: To investigate the effect of propolis on the proliferation and osteogenic differentiation of human periodontal ligament fibroblasts(HPDLFs) in vitro.Methods: HPDLFs were treated by various concentrations of the aqueous extract of propolis.The proliferation of HPDLFs was studied by MTT assay.The expressions of alkaline phosphatase(ALP) and osteocalcin(OC) of the cells were examined by immuno-enzyme assay and radio-immunologic assay respectively.Results: The aqueous extract of propolis at concentrations of 10- 150 μg/ml promoted the proliferation( P < 0.05), at 10 -200 μg/ml increased osteocalcin secretion and alkaline phosphatase activity( P < 0.05 ) of HPDLFs, the concentration of 100 μg/ml showed the strongest effects (P < 0.01 ).Conclusion:Aqueous extracts of propolis can promote the proliferation and osteogenic differentiation of HPDLFs.%目的:探讨蜂胶水提液对人牙周膜成纤维细胞(HPDLFs)生长增殖、骨向分化的影响,寻找蜂胶促增殖及分化作用的最佳浓度.方法:将不同浓度的蜂胶水提液加入体外培养的HPDLFs中,设置阳性对照和空白对照,通过检测HPDLFs的增殖(MTT法)、碱性磷酸酶活性及骨钙素含量,观察蜂胶水提取液对HPDLFs的增殖和分化的影响.结果:10~150μg/ml浓度的蜂胶水提液对体外培养HPDLFs的增殖均有促进作用,100μg/ml浓度时促增殖作用最明显.10~200 μg/ml蜂胶水提液均可提高HPDLFs的碱性磷酸酶活性、促进HPDLFs的骨钙素合成(P<0.05),其中以100 μg/ml蜂胶水提液作用最显著(P<0.01).结论:低浓度的蜂胶水提液能促进HPDLFs增殖和向成骨细胞方向分化.
