Objective To detect the serum levels of ubiquitin (Ub) and 20S proteasome (20S PSM) in patients with chronic obstructive pulmonary disease (COPD), and their relationship with tumor necrosis factor-a(TNF-α). Methods Serum levels of Ub, 20S PSM, TNF-a were determined by enzyme-linked immunosorbent assay in 30 patients with COPD in acute exacerbation (COPD I group), 30 patients with COPD in remission (COPD Ⅱ group), and 30 healthy subjects (control group). Results As compared with control group and COPD II group, serum levels of Ub, 20S PSM, and TNF-a were significantly increased (P < 0.01) in COPD Ⅰ group. Serum levels of Ub and TNF-α were statistically increased in COPD II group as compared to control group (P < 0.01), while 20S PSM was not significantly increased. A significant correlation was found between serum levels of Ub, 20S PSM, and TNF-α. Conclusion Serum levels of Ub and 20S PSM were increased, may be related to high protein catabolism, and was associated with systemic inflammatory response in patients with COPD.%目的:检测慢性阻塞性肺疾病(COPD)患者血清泛素(Ub)、20S 蛋白酶体(20S PSM)的变化,并探讨其与肿瘤坏死因子-α(TNF-α)的相关性.方法:采用酶联免疫吸附测定法检测30 例COPD 急性加重期患者(COPDⅠ组)、30 例COPD 缓解期患者(COPDⅡ组)及30 例健康体检者(正常对照组)血清Ub、20S PSM、TNF-α水平,并进行分析.结果:与正常对照组及COPDⅡ组比较,COPDⅠ组血清Ub、20S PSM、TNF-α均显著增加(P < 0.01).与正常对照组比较,COPDⅡ组血清Ub、TNF-α显著增加(P < 0.01),20S PSM 增加不显著(P > 0.05).COPD 患者血清Ub、20S PSM 水平分别与TNF-α具有相关性(r = 0.559,r = 0.859,均P < 0.01).结论:Ub、20SPSM 在COPD 患者血清中含量增加,可能与COPD 患者体内蛋白质高分解代谢相关,并且其水平增加与COPD全身炎症反应因子TNF-α激活相关.
