HMGA2和E-Cadherin在前列腺癌组织中的表达及意义

摘要

目的 探讨高迁移率蛋白A2(HMGA2)和上皮型钙黏蛋白(E-Cadherin)在前列腺癌侵袭、转移中的作用.方法 选取2008-2016年于大连市中心医院手术切除的60例前列腺癌石蜡包埋标本,以癌旁正常前列腺组织作对照(对照组),采用EliVision两步法检测HMGA2和E-Cadherin在前列腺癌中的表达,并将检测结果结合患者的临床资料进行分析.结果 前列腺癌中HMGA2的阳性表达率高于对照组(χ2=51.818,P<0.01).前列腺癌中E-Cadherin的阳性表达率低于对照组(χ2=53.494,P<0.01).肿瘤伴有转移,侵犯精囊腺,Gleason分级>7分,Ⅲ~Ⅳ期前列腺癌组,HMGA2的阳性表达率增高,差异有统计学意义(P<0.01);而E-Cadherin的阳性表达率降低,差异有统计学意义(P<0.05).前列腺癌中HMGA2与E-Cadherin的阳性表达具有相关性(r = -0.569,P < 0.01).结论 前列腺癌中HMGA2的高表达和E-Cadherin的低表达与肿瘤的侵袭和转移有关,两者之间具有协同作用.%Objective To investigate the role of high mobility group protein2(HMGA2)and E-Cadherin in the development of prostate cancer(PCa). Methods A total of 60 paraffln-embedded specimens of PCa were collected from patients who underwent operation in Dalian Central Hospital from 2008 to 2016.The expression lev-els of HMGA2 and E-Cadherin in the specimens were examined by immunohistochemical staining.The correlations between the expression of HMGA2 and E-Cadherin and clinical features such as age,Gleason score,invasion,me-tastasis and TNM stage were studied. Results The expression levels of HMGA2 in PCas were significantly higher than that in controls(χ2=51.818,P<0.01),but the positive expression levels of E-Cadherin in PCas were lower than that in controls(χ2=53.494,P<0.01).The positive expression of HMGA2 and the loss of E-Cadherin were confirmed in PCa accompanied by metastasis,seminal invasion,Gleason score>7 and stageⅢ~Ⅳ(P<0.05). Overexpression of HMGA2 was associated with down-regulation of E-cadherin(Spearman′s r=-0.569,P<0.01). Conclusions Up-regulation of HMGA2 and down-regulation of E-Cadherin are cooperatively correlated with the invasion and metastasis in PCa.
