凝胶过滤层析参数对EV71病毒粗提液分离效果的影响

摘要

通过凝胶过滤层析达到分离纯化EV71肠道病毒粗提液,以提高EV71的纯度.选用不同的洗脱流速、不同样品上样量、不同的缓冲溶液的pH值和不同盐浓度对EV71肠道病毒粗提液进行连续洗脱.结果表明,凝胶过滤层析的洗脱流速、样品上样量、缓冲溶液的pH值和盐浓度对EV71病毒均能不同程度地影响EV71病毒粗提液的分离效果.在试验范围内,选择洗脱流速为60 cm.h-1、样品的上样量为5％ CV、缓冲溶液的pH为7.5和盐浓度为0.1 mol·L-1时,对EV71病毒粗提液的分离效果比较理想.%The influence of gel-filtration chromatography flow rate, the sample weight, buffer pH value and salt concentration on separation performance of the protein crude extracts of EV71 were investigated. The results showed that all the above four factors could affect the separation performance of the crude extracts in gel-filtration chromatography by various degrees. In the scope of the present tests, the best separation performance was achieved using 60 cm·h-1 rate,5%CV sample, pH 7.5 buffer and with 0.1 mol·L-1 salt concentration.
