蛋氨酸脑啡肽对GM-CSF诱导的小鼠骨髓来源树突状细胞TLR-4和TNF-α表达影响

         

摘要

To observe the influence of Methionine enkephalin (MENK)on the expression of toll like receptor 4(TLR-4) and tumor necrosis factor-α(TNF-α) of DCs derived from murine bone marrow progenitors (BM-DCs)induced by MENK and to investigate the immunity regulatory mechanism of MENK on DCs.The BM-DCs were prepared and cultured in vitro,induced by GM-CSF and MENK.The mRNA expression of TLR-4 and TNF-α were detected by RT-PCR.The protein expression of TLR-4 and TNF-α were detected by ELISA and Western-Blot technique respectively after seven days.The expression of TLR-4 and TNF-α in MENK group were both increased compared to normal control group (P<0.01) and GM-CSF group (P<0.05).This suggested that MENK is probably to promote the expression of TLR-4 and TNF-α from gene and protein level for DCs.%观察蛋氨酸脑啡肽(MENK)对巨噬细胞-粒细胞集落刺激因子(GM-CSF)诱导的小鼠骨髓来源树突状细胞TLR-4和TNF-α表达的影响,探讨蛋氨酸脑啡肽对小鼠骨髓来源树突细胞免疫功能的调节机制.体外制备小鼠骨髓细胞用GM-CSF协同MENK诱导培养7d后用RT-PCR法检测TLR-4和TNF-α的mRNA表达,用Western-Blot和ELISA法分别检测TLR-4和TNF-α蛋白的表达.结果表明,MENK协同诱导组TNF-α和TLR--4表达高于正常对照组(P<0.01)和GM-CSF诱导组(P<0.05).显示MENK可以促进树突状细胞TLR-4和TNF-α的表达.
