洋川芎内酯A对大鼠脑缺血再灌注损伤的保护作用研究

             

摘要

Objective:To investigate the effect of senkyunolide A (ASE) on cerebral ischemia-reperfusion injury (IRI) in rats and the related mechanism. Methods:40 Sprague-Dawley male rats were randomly divided into 4 groups:sham-operation group(Sham,n=15), model group (CTL, n=15) and and treatment group treated with ASE (ASE-H group and ASE-L group). Middle cerebral artery was occluded by thread for 90min, followed by reperfusion. The neurological outcomes including edema index, infare size and neurological deficit scores were evaluated 24h after MCAO. The levels of interleukin (IL)-1β, IL-6 and malonaldehyde (MDA), and activity of superoxide dismutase (SOD) in the brain tissue were assessed according to the corresponding kits. Results:As compared with CTL group, ASE significantly reduced the edema index, infarc size and neurological deficit scores, and the reduction of ASE-H group is obviously better than ASE-L group (P<0.05). Also, ASE decreased the tissue levels of IL-1β, IL-6, and MDA, and enhanced the activity of SOD compared to CTL group, and the difference was statistically significant (P<0.05). Conclusion ASE can protect against focal brain IRI partly through the mechanism of attenuating oxidative stress and inhibiting the over-expression of pro-inflammatory cytokines.%目的:探讨洋川芎内酯A对大鼠脑缺血再灌注损伤的保护作用及其机制。方法:将60只雄性健康SD大鼠随机分为4组,分别为:假手术组(Sham,n=15),模型组(CTL,n=15),洋川芎内酯A低剂量组(ASE-L,n=15)和洋川芎内酯A高剂量组(ASE-H,n=15),大鼠脑缺血90min后恢复血流,于大脑中动脉阻断(MCAO)后24h将大鼠处死,对各组脑组织中IL-1β、IL-6、MDA水平以及SOD活性进行测定,并对各组的缺血侧脑水肿指数、脑梗死体积以及神经功能评分进行比较。结果:与CTL组比较,给药组大鼠的脑水肿指数、脑梗死体积以及神经功能评分明显改善,高剂量组(ASE-H)明显优于低剂量组(ASE-L),差异均具有统计学意义(P<0.05);同时,给药组大鼠脑组织中IL-1β、IL-6、MDA水平明显降低,SOD活性明显提高,差异也具有统计学意义(P<0.05)。结论:围术期采用洋川芎内酯A预处理可明显减轻脑组织的氧化应激反应,抑制促炎因子释放,对于脑缺血再灌注引起的中枢神经系统损伤具有明显保护作用。
