芪桑方对2型糖尿病大鼠血糖、血脂及蛋白酪氨酸磷酸酶1B 表达的影响

摘要

Objective To observe the influence of qisang fang on blood glucose ,serum lipoprotein and protein tyrosine phosphatase 1B(PTP1B)expression in type 2 diabetes rats .Methods Type 2 dia‐betes model was induced with high sucrose-fat diet and streptozocin ,rats were randomly divided into groups :normal group ,model group ,metformin treatment group ,qisang fang low dose treatment group ,qisang fang middle dose treatment group ,qisang fang high dose treatment group .Blood glucose , serum lipoprotein ,hemoglobin Alc (HbAlc) and PTP1B expression in rats were detected after treat‐ment .Results Compared with normal group ,HbAlc levels were significantly higher( P <0 0.1) ,LDL , TG and TCH were significantly increased( P <0 0.5) ,HDL decreased( P <0 0.5) in model group .Qi‐sang fang can reduce the levels of TG ,TCH ,LDL ,FBG ,HbAlc ,improve the level of HDL ,and inhibit the PTP1B expression in liver .Conclusion Qisang fang can effectively improve sugar and lipid metab‐olism in T2DM rats ,inhibit the PT P1B expression may be the mechanism .%①目的观察芪桑方对2型糖尿病模型大鼠血糖、血脂的调节作用及其对蛋白酪氨酸磷酸酶1B（PTP1B）表达的影响。②方法采用高脂、高糖饲料加腹腔注射链脲佐菌素的方法进行2型糖尿病大鼠模型的造模。实验动物分为正常组、2型糖尿病模型组、二甲双胍阳性对照组、芪桑方低剂量治疗组、芪桑方中剂量治疗组、芪桑方高剂量治疗组。连续灌胃给药30d，测空腹血糖（FBG）、糖化血红蛋白（HbAlc）、总胆固醇（TCH）、甘油三酯（TG）、高密度脂蛋白（HDL）、低密度脂蛋白（LDL）及各组大鼠肝脏PTP1B的表达。③结果与正常组比较，模型组大鼠HbAlc含量显著增高（P＜00．1），TCH、TG、LDL明显升高（P＜00．5），HDL明显下降（P＜00．5）。芪桑方可降低2型糖尿病模型大鼠的FBG、HbAlc、TCH、TG、LDL水平升高，HDL水平与模型组比较有明显改善（P＜00．5），芪桑方可降低2型糖尿病大鼠肝中PTP1B的表达。④结论芪桑方能有效地改善2型糖尿病大鼠糖代谢和脂代谢，抑制PTP1B表达可能为其作用机制之一。
