邻苯二甲酸二(2-乙基已基)酯对雄性成年大鼠脑神经细胞的毒性作用

     

摘要

目的 观察邻苯二甲酸二(2-乙基已基)酯[di -(2- ethylhexyl)phthalate,DEHP]对成年雄性大鼠脑神经细胞的毒性作用.方法 将32只雄性健康成年SD大鼠随机分为溶剂对照组(玉米油)和DEHP 375、750、1500 mg·kg-1染毒组.采用经口灌胃的方式重复染毒30d,于末次染毒24h后,10%水合氯醛麻醉,取脑组织称重后进行病理形态学观察及Tunnel法检测神经细胞凋亡情况.结果 各DEHP染毒组大鼠脑脏器系数与玉米油组比较差异无统计学意义(P>0.05).病理形态学结果显示随DEHP染毒剂量增加,神经元细胞细胞核深染、不同程度固缩,核仁不清,染色质凝集,胞浆呈空泡样改变加重.DEHP染毒750、1500 mg·kg-1组与玉米油组比较大鼠皮质区神经细胞凋亡数差异具有统计学意义(P<0.01),1500 mg·kg -组与玉米油组比较海马区神经细胞凋亡数差异具有统计学意义(P<0.01).结论 DEHP经口短期重复染毒对成年雄性大鼠脑组织可能会产生毒性作用.%Objective To observe the toxicity of di - (2 -ethylhexyl) phthalate on nerve cells in adult male rats. Methods 32 adult male Sprague -Dawley(SD) rats were randomly divided into four groups, namely: the solvent control group (corn oil), DEHP poisonous group (375, 750, 1500 mg ? Kg-1 ). SD rats were given the different dose of DEHP by the way of gastric lavage through the mouth for 30 days, and at the last time, 24 hours later after administration, 10% hydration chlorine aldehyde mixture were used to anesthesia, weighing the brain, having the pathological morphology observation of brain and using the method of Tunnel to detect the apoptosis of the nerve cell. Results Compared with control group , Visceral index of brain in all DEHP groups had no statistically significant difference (P >0.05). Pathological morphology results showed with DEHP's dosage increased, Neurons with hyperchromatic nuclei, becoming pyknotic at Different degree, prominent nucleoli was not clear, chromatin condensed, The cytoplasm changed like empty bubble becoming heavier. Compared with control group , the number of apoptosis nerve cells of the brain cortex in 750,1500 mg ? Kg-1 DEHPgroup had statistically significant difference( P <0.01). The number of apoptosis nerve cell of the hippocampuss between 1500 mg/kg DEHP group and controls had statistically significant difference ( P < 0.01). Conclusion DEHP may produce the toxic effect on nerve cells by the way of short - term repeated exposure in adult male rats.
