同型半胱氨酸对血管平滑肌细胞增殖和细胞周期的影响及叶酸的拮抗作用

             

摘要

目的 探讨不同浓度同型半胱氨酸(Hcy)对血管平滑肌细胞(VSMCs)增殖和细胞周期分布的影响及叶酸对Hcy的拮抗作用.方法 正常人脐动脉VSMCs体外培养,随机分为6组:空白对照组、不同浓度Hcy干预组(50、100、200、500μmol·L-1)和叶酸治疗组(Hcy100μmol·L-1+叶酸30μmol·L-1).应用MTT法及流式细胞仪检测各组VSMCs增殖率及细胞周期.结果 随着Hcy浓度的增加VSMCs的增殖率增加;Hcy作用后VSMCs G0/G1期细胞数降低;S期和G2/M期细胞数增加,处于DNA合成期的细胞数明显增加,而叶酸治疗组与空白对照组比较差异无统计学意义(P>0.05),叶酸治疗组与Hcy 100μmol·L-1组比较差异有统计学意义(P<0.05).结论 Hcy可通过对细胞周期的影响促进VSMCs的增殖,并在一定时间和浓度范围内呈明显的剂量依赖关系;叶酸对Hcy有拮抗作用.%Objective To explore the effect of homocysteine ( Hey ) on the proliferation of vascular smooth muscle cells ( VSMCs ) and the antagonizing effect of folic acid. Methods Cultured VSMCs were randomly divided into black control group, different concentrations of Hey group ( 5(K10(K20(K500uM )and folic acid treated group( HcylOOuM + folic acid 30uM ). The effect of folic acid treated group and different concentrations of Hey group on the proliferation rate and cell cycle of VSMCs were detected by MTT and cell cycle flow cy-tometer. Results With Hey concentration increasing, the multiplication rate of the VSMCs increased; and the VSMCs in the G0/G1 phase decreased and in the S and G2/M phase also increaced. It meant that the VSMCs number in the synthesis period of DNA increased significantly. There were no significant changes between folic acid treated group and black control group. VSMCs between folic acid treated groupand Hey 100μmol ? L-1 group showed significant difference ( P <0. 05 ). Conclusion Hey can promote the proliferation of VSMCs in a manner of dose dependent and time dependent; folic acid has the antagonistic effects on Hey.
