槐定碱单分散微粒的制备与药物缓释

         

摘要

Objective To prepare monodisperse sophoridine-loaded microparticles and to investigate the influence of particle size,PLGA with different molecular weight(MW)and monomer ratio(L/G),different solvents on sophoridine-loaded microparticles during the hydrogel template process.Methods Sophoridine-loaded microparticles were fabricated by a novel approach of hydrogel template method,and were then analyzed on morphology,drug loading and encapsulation efficiency,and in vitro release profiles.Results The obtained microparticles exhibited as microdiscs with homogeneous size and predefined shape. The release profiles microspheres was closely related to its particle size,PLGA with different MW and L/G,and solvent systems with low drug loading capacity.Conclusion The hydrogel template technique can be applied to precisely define the size and shape,and are suitable to different solvents,which is beneficial for targeted drug delivery system of microparticles.%目的 制备槐定碱单分散缓释微粒,考察凝胶模板制备法中微粒粒径大小、聚乳酸-羟基乙酸共聚物[poly(lactic-co-glycolic acid),PLGA]种类和不同溶剂体系对槐定碱微粒缓释属性的影响.方法 用凝胶模板法制备不同种类槐定碱-PLGA微粒,显微观察表面形态,测定载药量及包封率,比较体外释药特征.结果 所制微粒呈微小圆柱体,粒径均一、形态可控;载药量与包封率均较低;微粒大小、PLGA种类及溶剂体系对微粒体外释药行为有明显影响.结论 凝胶模板制备技术能调控微粒粒径、形态,适用于槐定碱单分散缓释微粒的制备.
