目的：探讨高效液相色谱法（ HPLC）测定盐酸法舒地尔含量的可行性。方法采用高效液相色谱法测定自制盐酸法舒地尔含量。色谱柱：phecda C18（250 mm ×4．6 mm，5μm），以1．0％三乙胺水溶液（用磷酸调节pH值至7．0）-甲醇（50∶50）为流动相；检测波长为275 nm，流速：1．0 ml/min；进样量：20μl；柱温：25℃。结果 HPLC法测定盐酸法舒地尔的浓度在0．0484～0．2420 g/L的范围内，浓度与峰面积有良好的线性关系（r＝0．9994，n＝5），平均回收率为（99．4±0．47）％， RSD为0．47％（ n＝9）。结论 HPLC法测定简单，专属性强、灵敏度高，可以用于盐酸法舒地尔的含量测定。%Objective To investigate the feasibility of determination of fasudil hydrochloride content by high performance liq -uid chromatography(HPLC).Methods HPLC was used to determine the content of a self-prepared fasudil hydrochloride solution . Separation was performed on the phecda C 18 column (250 mm ×4.6 mm, 5 μm) with the mobile phase of triethylamine solution (the pH value adjusted to 7.0 using phosphoric acid)-methanol (50∶50), at a detection wave length of 275 nm, a flow rate of 1.0 ml/min, feed volume of 20μl and column temperature of 25℃.Results With HPLC, the content level was within 0.0484-0.2420 g/L. Linear relationship could clearly be noted between the content and the peak area (r=0.9994, n=5).Average recovery rate was (99.4 ± 0.47)%and relative standard deviation (RSD) was 0.47%(n=9).Conclusion HPLC was a simple, sensitive and specific detec-tion method, which could be used for the determination of fasudil hydrochloride content .
