目的:探讨高效液相色谱法( HPLC)测定盐酸法舒地尔含量的可行性。方法采用高效液相色谱法测定自制盐酸法舒地尔含量。色谱柱:phecda C18(250 mm ×4.6 mm,5μm),以1.0%三乙胺水溶液(用磷酸调节pH值至7.0)-甲醇(50∶50)为流动相;检测波长为275 nm,流速:1.0 ml/min;进样量:20μl;柱温:25℃。结果 HPLC法测定盐酸法舒地尔的浓度在0.0484~0.2420 g/L的范围内,浓度与峰面积有良好的线性关系(r=0.9994,n=5),平均回收率为(99.4±0.47)%, RSD为0.47%( n=9)。结论 HPLC法测定简单,专属性强、灵敏度高,可以用于盐酸法舒地尔的含量测定。%Objective To investigate the feasibility of determination of fasudil hydrochloride content by high performance liq -uid chromatography(HPLC).Methods HPLC was used to determine the content of a self-prepared fasudil hydrochloride solution . Separation was performed on the phecda C 18 column (250 mm ×4.6 mm, 5 μm) with the mobile phase of triethylamine solution (the pH value adjusted to 7.0 using phosphoric acid)-methanol (50∶50), at a detection wave length of 275 nm, a flow rate of 1.0 ml/min, feed volume of 20μl and column temperature of 25℃.Results With HPLC, the content level was within 0.0484-0.2420 g/L. Linear relationship could clearly be noted between the content and the peak area (r=0.9994, n=5).Average recovery rate was (99.4 ± 0.47)%and relative standard deviation (RSD) was 0.47%(n=9).Conclusion HPLC was a simple, sensitive and specific detec-tion method, which could be used for the determination of fasudil hydrochloride content .
