HPLC-ELSD 法测定九味肾清颗粒中黄芪甲苷的含量

摘要

目的：建立检测九味肾清颗粒中黄芪甲苷含量的高效液相色谱-蒸发光散射（HPLC-ELSD）法。方法以黄芪甲苷为测定指标，色谱柱为依利特 Hypersil ODS 5μm，4．6 mm×250．0 mm，流动相为乙腈-水（38∶62）；流速1．0 mL·min－1，漂移管温度110℃；载气流量2．59 L·min－1。结果黄芪甲苷对照品线性范围为1．7588～17．5880μg，回归方程为 Y ＝1．3528X ＋4．7859，r ＝0．9997，平均回收率为101．51％，RSD 为2．91％。结论HPLC-ELSD 法可以准确地进行定量分析，可用于九味肾清颗粒的质量控制。%Objective To establish a high performance liquid chromatography-evaporative light scattering detection (HPLC-ELSD)method for determining the content of astragaloside in Shenq-ing granules.Methods The HPLC-ELSD of astragaloside was performed on the Hypersil ODS chromatography column (4.6 mm×250.0 mm,5 μm)with acetonitrile-water (38∶62)at a flow rate of 1.0 mL·min-1 .The drift tube temperature was set at 110 ℃,with a carrier gas flow rate of 2.59 L· min-1 .Results The linear range of astragaloside reference substance was 1.758 8-17.588 0 μg.The re-gression equation wasY=1.352 8X+4.785 9 (r=0.999 7).The average recovery rate was 101.51%, with a RSD of 2.91%.Conclusion The HPLC-ELSD method is accurate for quantitative analysis of astragaloside and can be used for controlling the quality of Shenqing granules.
