矢车菊黄素诱导 A549细胞凋亡及机制研究

摘要

目的：研究矢车菊黄素对 A549细胞增殖及凋亡的影响及其机制。方法采用 MTT 法测定 A549细胞增殖，Hochest 染色检测细胞凋亡，流式细胞仪分析细胞凋亡率及细胞周期，Western blot 方法检测细胞中 NF-κB P65蛋白的表达。结果矢车菊黄素剂量依赖性抑制 A549细胞的增殖，半数抑制浓度（IC50）值为0．67μmol·L－1；剂量依赖性诱导 A549细胞凋亡，0．15、0．30μmol·L－1矢车菊黄素处理时诱导细胞发生 G1期阻滞；剂量依赖性上调 A549细胞中 NF-κB P65的表达。结论矢车菊黄素能诱导 A549细胞的凋亡，其促凋亡作用可能与 NF-κB 信号通路激活有关。%Objective To investigate the effect of centaureidin on the proliferation and apopto-sis of A549 cells and its mechanism of action.Methods The proliferation of A549 cells was meas-ured by MTT assay,the apoptosis by Hochest staining,the apoptosis rate and cell cycle by flow cytometry,and the expression of NF-κB P65 by Western blot.Results Centaureidin treatment suppressed proliferation,induced apoptosis and up-regulated NF-κB P65 expression in A549 cells in a dose-dependent manner.The median inhibitory concentration (IC50 )was 0.67 μmol·L-1 .G1 phase arrest was found in A549 cells treated with centaureidin at a concentration of 0.15 or 0.30 μmol·L-1 .Conclusion Centaureidin can induce apoptosis in A549 cells via activating NF-κB signal pathway.
