目的 探索人胚干细胞来源的成纤维细胞(EBF)作为纳米银颗粒细胞毒性评价模型的可行性,旨在为纳米材料生物安全性评价提供新的细胞模型.方法 将纳米银与EBF细胞共孵育,采用相差显微镜、CCK-8法、流式细胞术和透射电子显微镜分别对细胞形态、细胞增殖、细胞周期及细胞摄取进行研究.结果 纳米银颗粒对EBF活性的影响是浓度和时间依赖性的,纳米银可以进入细胞内并引起细胞周期阻滞.结论 EBF可以作为一种新的纳米银颗粒生物安全性评价模型,由于其来源的特殊性,可进一步模拟人体正常细胞的反应.%Objective To explore the feasibility of human embryonic stem cell-derived fibroblasts (EBF) to be as a novel cell model to evaluate the cytotoxicity of silver nanoparticles (Ag-NPs). Methods After exposing to Ag-NPs the cell morphology, cell proliferation, cell cycle and cellular uptake were examined by phase-contrast microscope, CCK-8, flow cytometry and transmission electron microscope. Results The cytotoxicity of Ag-NPs in EBF was dose and time-depedent. Ag-NPs can enter the cells and induced a G2/M arrest. Conclusion EBF could be taken as a valid candidate for cytotoxicity screening assays of nanoparticles as this model achieves a closer representation of the in vivo response.
