目的:观察阿魏菇多糖(polysaccharides from Pleurotus ferulae,PFP)、顺铂(DDP)及两者联合对小鼠宫颈癌细胞株U14移植瘤的抑制作用及凋亡诱导作用,并初步探讨阿魏菇多糖的抗肿瘤作用机制.方法:建立昆明小鼠U14宫颈癌模型,并随机分为四组:模型对照组、PFP组、DDP组、PFP+DDP组,治疗10天后检测各组瘤重及抑瘤率,采用TUNEL法检测各组瘤组织凋亡指数(apoptosis index,AI).结果:模型组小鼠体内瘤体积、瘤重明显高于各干预组(P<0.01),PFP+DDP组小鼠体内瘤重和瘤体积分别为[(0.40±0.61)g,(197.84±11.99)mm3 ]低于DDP组[(0.61±0.26)g,(211.46±14.58)mm3]; PFP组、DDP组、PFP+ DDP组瘤组织凋亡指数均高于模型对照组,其中以PFP+DDP组最明显.结论:PFP联合DDP比两药单独应用抑制肿瘤生长及诱导肿瘤细胞凋亡的作用更强,二者具有协同抗癌作用.PFP可以增强DDP的药物敏感性,其作用机制可能是通过诱导宫颈癌细胞凋亡而实现的.%Objective :To observe the growth - inhihition and the apoptosis induced hy polysaccharides from Pleurotus ferulae ( PFP ), cisplatin( DDP ) and their combination on murine cervical carcinoma U14; To investigate the mechanisms of PFP in the treatment of cancer. Methods: The mice with U14 were randomly divided into four groups : Model group , DDP group , PFP group , PFP + DDP group. After 10 days , the inhihitory rates of each group were detected. The U14 cell apoptosis index ( AI ) was assessed by TUNEL. Results : The weight and volume of tumor in model group were higher than that in other three groups( P <0. 01 ). The weight and volume of tumor in PFP + DDP group ( 0. 40 ±0. 61 )g and ( 199. 82 ± 13. 62 )mm3 , respectively , were lower than that in DDP group ( 0. 61 ±0. 26 ) g and ( 209. 51 ±23. 28 )mm3. The tumor cell apoptosis in experimental group was higher than those in model group, and the apoptosis index was highest in PFP + DDP group. Conclusion: PFP and DDP comhined modality therapy could markedly inhibit the growth of the transplantation tumor of murine cervical carcinoma U14 and induce the tumor cell apoptosis, the effect was more strong than PFP, DDP. PFP could enhance the sensitivity of cervical carcinoma to DDP.
