血小板生成素对巨核细胞的增殖及保护作用体外研究

             

摘要

目的:观察巨核细胞中加入血小板生成素(thrombopoietin,TPO)之前和加入之后卡铂对其影响的变化,以及TPO对巨核细胞的促增殖作用,从而了解TPO对MO7e细胞的增殖及保护作用.方法:MTT法测定不同浓度的TPO对巨核细胞株MO7e作用24h、48h、72h的影响,作出增殖曲线.测定不同浓度卡铂对加入TPO前、后的MO7e细胞作用24h、48h的抑制率,求出半数抑制浓度(IC50),比较两者的抑制率和IC50.结果:MO7e细胞增殖率随TPO浓度增大而增大,TPO浓度为256ng/ml时,进入平台期.加入TPO后,卡铂对MO7e细胞的抑制率明显降低,IC50约为加入TPO前的2倍.结论:TPO可以促进巨核细胞增殖,该作用呈剂量依赖性,同时可减弱巨核细胞被化疗药物抑制,说明对巨核细胞有保护作用.%Objective;To research what change will occur over the effect of carboplatin on M07e cell alone and on MO7e cell with the addition of thrombopoietin(TPO) , and the proliferation effect of TPO on megakaryocyte, as a result , having an awareness of the effect of TPO on M07e cell in proliferation and protection. Methods: MTT assay was employed to delect the different concentration of TPO plays a role on megakaryocyte cell line, M07e cell, respectively during 24h, 48 h, 72h and make a proliferation curve concerned. And to detect the inhibition ratio over different concentration of carborplatin to M07e cell and to M07e cell with the addition of TPO, respectively during 24h and 48h. Their half inhibition concentration (ICW) as well as inhibition can be worked out and compared. Results: M07e cell proliferation rate increased as the concentration of TPO increased. When TPO concentration was 256ng/ml the rate got to plateau. After the addition of TPO, the inhibition ratio of carborplatin to M07e cell significantly decreased and the ICM was 2 times as much as the IC^, without TPO. Conclusion: TPO can promote the proliferation of megak-eryocyties, which relies on the dose. Simultaneously, TPO can, to a greater degree, protect megakeryocyties against the inhibition from chemotherapy drugs.
