肺癌细胞中 RASSF4的高表达抑制肺癌细胞的增殖和侵袭能力

摘要

目的：探讨 RASSF4在肺癌中的增殖和侵袭能力。方法：向肺癌细胞系 H460和 A549中导入RASSF4的 cDNA 质粒后，通过 MTT、克隆形成实验、基质胶侵袭实验、流式细胞术等方法检测肺癌细胞的生长和侵袭能力。结果：RASSF4能够通过下调 MMP2、MMP9和 cyclinD1的表达，抑制肺癌细胞的侵袭、增殖及克隆形成能力。结论：RASSF4在肺癌细胞中通过抑制细胞生长及侵袭能力发挥重要的肿瘤抑制功能。%Objective:Despite the results showed that RASSF4 play the tumor suppressor role in tumor cells,no studies investigating its role in human non - small cell lung cancer( NSCLC). Methods:In the present study, RASSF4 plasmid transfection was performed in H460 and A549 cell lines. The cell growth was observed using a3 -(4,5 - dimethylthiazol - 2 - yl)- 2,5 - diphenyltetrazolium bromide(MTT)assay,flow cytometry analysis and colo-ny formation. The invasive ability of cell was observed by Matrigel invasion assay. Results:RASSF4 overexpression in-hibited proliferation,colony formation,and invading ability with downregulation of cyclin D1,MMP2 and MMP9. Con-clusion:RASSF4 was a tumor suppressor in NSCLC.
