奈达铂对喉癌细胞系放射敏感性的影响及机制

     

摘要

目的:探讨奈达铂对喉癌细胞系(Hep-2细胞)放射敏感性的影响并初步探讨其机制.方法:设空白组、低浓度奈达铂组、高浓度奈达铂组、空白+照射组、低浓度奈达铂+照射组、高浓度奈达铂+照射组,单次照射6Gy,流式细胞仪检测细胞凋亡及RT-PCR检测血管内皮生长因子(VEGF)水平并半定量VEGF以初步探讨机制.结果:奈达铂对喉癌Hep-2细胞生长有抑制作用.空白组、低浓度奈达铂组、高浓度奈达铂组细胞的D0值分别为1.98、1.32、0.86Gy,Dq值分别为1.99、1.27、1.02Gy,α值分别为0.113、0.173、0.536(Gy-1)和β值分别为0.105、0.260、0.521(Gy-2).空白组、低浓度奈达铂组、高浓度奈达铂组、空白+照射组、低浓度奈达铂+照射组、高浓度奈达铂+照射组的细胞凋亡率分别为1.20%、1.15%、2.18%、2.65%、2.99%、4.98%.高浓度奈达铂组、空白组+照射组、低浓度奈达铂+照射组及高浓度奈达铂+照射组VEGF的表达水平较空白组及低浓度奈达铂组低,且高浓度奈达铂+照射组表达最低.结论:奈达铂对人喉癌细胞有放射增敏作用,其机制可能与降低VEGF表达和促进细胞凋亡有关.%Objective:To investigate the effects of Nedaplatin on the radiosensitivities of human laryngeal carcino-ma and the mechanism. Methods:The radiosensitivity of human laryngeal carcinoma cells transfected and untransfect-ed with Nedaplatin were studied by cell survival curve which protracted by clonogenic assay and caculating D0、Dq . Fi-nally we investigated the correlated mechanism throw the cell apoptosis which studied by flow cytometry and the VEGF content tested by RT-PCR. Results:The radiosensitivity of human laryngeal carcinoma transfected with Nedaplatin was intensified and the D0 and Dq was lowered andαwas increased. The results of flow cytometry showed that the cell apoptosis rate with Nedaplatin and radiotherapy were increased. The VEGF content of human laryngeal carcinoma transfected with Nedaplatin and after radiotherapy respectively was decreased. Conclusion:Nedaplatin could intensify the radiosensitivity of human laryngeal carcinoma cell. Nedaplatin and radiotherapy could decrease the VEGF content of human laryngeal carcinoma cell.
