ELL2在人成骨肉瘤中的表达及对MG-63细胞增殖的影响

Objective:To explore the expression of ELL2 in human osteosarcoma and check its impact on proliferation of osteosarcoma cell line.Methods:RT-PCR was used as detect the expression of ELL2 in human osteosarcoma tissues and normal adjacent tissues.RT-PCR and Western-blot were uses as detect the expression of ELL2 in human osteosarcoma cell lines and normal osteoblast cell line.MTT assay and cell count assay were utilized to study the influence of ELL2 overexpression on cell proliferation.Results:ELL2 was down-regulated in osteosarcoma tissues than normal adjacent tissues (P=0.001).ELL2 was also down-regulated in osteosarcoma cell lines than normal osteoblast cell line (P<0.05).Overexpression ELL2 in MG-63 cell line inhibited its proliferation (P<0.05).Conclusion:ELL2 was down-regulated in osteosarcoma an ELL2 overexpression inhibits the proliferation of osteosarcoma cells.%目的:研究ELL2在人成骨肉瘤中的表达情况以及其对人成骨肉瘤细胞增殖的影响.方法:运用RT-PCR检测ELL2在人成骨肉瘤组织及其正常毗邻组织中的表达,RT-PCR及Western-blot检测ELL2在人成骨肉瘤细胞系及人正常成骨细胞系中的表达.构建ELL2过表达载体,MTT和细胞计数试验检测过表达ELL2对人成骨肉瘤细胞系MG-63增殖的影响.结果:ELL2在人成骨肉瘤组织中的表达显著低于其正常毗邻组织(P=0.001).ELL2在人成骨肉瘤细胞系中的表达相比于人正常成骨细胞系明显下调(P<0.05).过表达ELL2能够显著抑制MG-63细胞的增殖(P<0.05).结论:ELL2在人成骨肉瘤中表达下调,其过表达能够抑制肿瘤细胞的增殖.
