长链非编码RNATUG1在膀胱尿路上皮癌中的表达及对细胞增殖和凋亡的作用

         

摘要

目的:探讨长链非编码RNA牛磺酸上调基因1(taurine up-regulated gene 1,TUG1)在膀胱尿路上皮癌(bladder urothelial carcinoma,BUC)中的表达,及其对BUC细胞增殖和凋亡的调控作用.方法:实时定量PCR法检测TUG1基因在62例BUC组织和T24、J82细胞系中的表达.统计分析TUG1基因在BUC组织的表达与BUC临床病理因素的相关性.TUG1抑制剂smart silencer-TUG1(ss-TUG1)沉默T24和J82细胞中TUG1的表达.CCK8法检测T24和J82细胞的增殖活力.流式细胞术检测T24和J82细胞的凋亡率.结果:与癌旁组织及人膀胱上皮永生化细胞系SV-HUC-1相比,TUG1基因在BUC组织及T24、J82细胞系中的表达显著上调.随着BUC病理分级和分期的增加,BUC组织中TUG1基因表达明显增加.转染ss-TUG1能够显著抑制T24和J82细胞中TUG1的表达,TUG1沉默能够显著抑制T24和J82细胞的增殖活力,并促进其细胞凋亡.结论:长链非编码RNA TUG1在BUC组织及细胞系中高表达,其表达沉默能够抑制BUC细胞的增殖活力并诱导细胞凋亡.%Objective:To investigate the expression of long non-coding RNA(lncRNA)taurine up-regulated gene 1(TUG1)in bladder urothelial carcinoma(BUC)and its regulation on the proliferation and apoptosis of BUC cells.Methods:The expression of TUG1 gene in 62 BUC tissues and T24 and J82 cell lines was detected by real-time quantitative PCR.The correlation between the expression of TUG1 gene in BUC tissue and the clinicopathological factors of BUC was statistically analyzed.The expression of TUG1 in T24 and J82 cells was silenced by the TUG1 in-hibitor(smart silencer-TUG1,ss-TUG1).CCK8 assay was used to detect the proliferation of T24 and J82 cells,and flow cytometry was used to detect the apoptosis rate of T24 and J82 cells.Results:Compared with the paracancer-ous tissues and human bladder epithelial immortalized cell line SV-HUC-1,the expression of TUG1 gene was up-regulated in BUC tissues and T24,J82 cell lines.With the increasing of BUC pathological grade and stage,the expres-sion of TUG1 gene in BUC tissues was increased significantly.Transfection of ss-TUG1 significantly inhibited the ex-pression of TUG1 in T24 and J82 cells,and TUG1 silence significantly inhibited the proliferation of T24 and J82 cells,and promoted their apoptosis.Conclusion:Long non-coding RNA TUG1 is highly expressed in BUC tissue and cell lines,and its expression silencing can inhibit the proliferation and induce apoptosis of BUC cells.
