胰腺癌特异相关长链非编码RNA靶基因的筛选验证及分析

摘要

目的：筛选并明确胰腺癌特异相关长链非编码RN A （lncRN A ）在胰腺癌及癌旁组织中的表达差异，并对其进行初步分析。方法：采用实时定量逆转录聚合酶链反应（RT‐qPCR）检测筛选验证胰腺癌特异相关长链非编码 RNA MAP3K14、PPP3CB、DAPK13种靶基因在60例胰腺癌组织及配对癌旁组织表达差异最强的目标lncRNA ，为胰腺癌特异相关lncRNA的功能研究做铺垫。结果：长链非RNA MAP3K14，PPP3CB ，DAPK1在各自的胰腺癌及癌旁组织中的表达水平均有意义（ P＝0．000，t＝17．32；P＝0．000，t＝15．045；P＝0．013，t＝34．55）；在胰腺癌组织的表达量两两比较有差异（ P＜0．01，F＝32．37），三组基因在胰腺癌旁组织的表达水平两两比较差异显著（ P＜0．01，F＝296．6），其中DAPK1基因在胰腺癌及癌旁基因中表达量最高。结论：三种长链非编码RNA在胰腺癌及癌旁组织中明确表达，其中DAPK1基因表达量最高，说明DAPK1与胰腺癌的发生发展有关，且在胰腺癌组织中表达低于癌旁组织，说明DAPK1是胰腺癌一种可能的抑癌基因。%Objective :To screen and identify the differentially expressed of specific long non‐coding RNA relevant tar‐get genes of pancreatic cancer between pancreatic cancer tissues and paracancerous tissues ,and analysis it preliminary . Methods :To screen the differentially expressed of specific long non‐coding RNA MAP3K14 ,PPP3CB ,DAPK1 of pan‐creatic cancer between pancreatic cancer tissues and paracancerous tissues in 60 cases by RT‐qPCR ,which was prepared for the functional study of pancreatic cancer specific relevant lncRNA .Results:The expression levels of MAP3K14 , PPP3CB ,DAPK1 were significance between pancreatic cancer tissues and paracancerous tissues respectively ( P=0.000 ,t=17 .32 ;P=0 .000 ,t=15 .045;P=0 .013 ,t=34 .55);the expression of three genes are different in pancreatic tissue ,(P<0 .01 ,F=32 .37) ,the expression of three sets of gene in adjacent pancreatic tissue of pairwise comparisons were significant (P<0 .01 ,F=296 .6) ,which the expression of DAPK1 was highest in pancreatic cancer and paracan‐cerous tissues .Conclusion :Three long non‐coding RNAs in pancreatic cancer and adjacent pancreatic cancer tissue were expressed clearly ,which DAPK1 expressed highest ,this shows that DAPK1 was related to the occurrence and devel‐opment of pancreatic cancer ,and the expression level of DAPK1 in pancreatic cancer tissues was lower than that in the adjacent tissues ,indicating that DAPK1 is a possible tumor suppressor genes of pancreatic cancer .
