Objective To investigate the expression and function of Fcareceptor on human fibroblast - like synoviocyles (FLS) of rheumatoid arthritis ( RA). Methods RT - PCR and immunofluorescence technique were applied to detect the expression of Fcareceptor on human RA FLS. The effect of IgA on the level of interleukin - 6 (IL - 6) secreted by human RA FLS was measured by using ELISA. Results The results from RT - PCR and immunofluorescence staining confirmed the expression of Fcareceptor on human RA FLS. Compared with control, secretory IgA (slgA) increased the level of IL-6 released by human RA FLS after 12h incubation (P <0.01). Mon-omeric IgA (mlgA) did not result in significant changes on the IL-6 production of human RA FLS (P>0.05). After 12h incubation, sl-gA + TNF - a increased the IL-6 concentration dramatically, compared to slgA or TNF - aalone ( P < 0.01). Conclusion Fcareceptor is expressed on human RA FLS. SlgA increases the IL - 6 concentration of human RA FLS. SlgA and TNF - atogether have synergistic effect on the IL-6 production.%目的 探讨Fcα受体在类风湿性关节炎人成纤维样滑膜细胞的表达特征及其功能.方 法采用RT-PCR和免疫荧光技术,从基因和蛋白水平检测Fcα受体在类风湿性关节炎人成纤维样滑膜细胞(RA FLS)的表达;采用ELISA方法,检测lgA对RA FLS分泌IL -6的影响.结果 采用RT -PCR和免疫荧光染色方法,均检测到Fcα受体在RA FLS的表达.sIgA与RAFLS孵育12h后,细胞分泌IL -6的水平明显增高,差异具有显著性(P<0.01).mIgA与RA FLS孵育12h后,细胞分泌IL -6的量无明显改变(P>0.05).sIgA +TNF-α同时与RA FLS孵育12h后,细胞分泌IL -6含量明显高于sIgA或TNF -α单一组(P<0.01).结论 Fcα受体在类风湿性关节炎人成纤维样滑膜细胞存在表达,sIgA增高RA FLS分泌IL -6的水平,并与TNF -α产生协同作用.
