辣椒疫霉菌拮抗真菌M-12发酵条件的优化∗

         

摘要

The strain of M-12 isolated from soil, Pinicielium sp., had obvious antimicrobial activa⁃tion against Phytophthora capsici. The antifungi test was studied on the medium technique with fungi⁃cides. The single factor experiment and the orthogonal test method were used to optimize the nutrition conditions and fermentation conditions. The results showed that the optimal nutritional components included corn flour 40 g/L, sucrose 40 g/L, peptone 3 g/L, KCl 0�5 g/L, MgSO4 ·7H2 O 0�5 g/L , K2HPO41�0 g/L and FeSO4·7H2O 0�01 g/L. The optimal fermentation conditions were medi⁃um volume in the flask 125 mL/250 mL, initial pH 6�0,inoculation dose 6%, seed age 60 h, fer⁃mentation temperature 26℃, table concentrator rotatory velocity 175 r/min and fermentation period 8 days.%采用土壤中分离筛选出的拮抗辣椒疫霉菌的青霉属菌株M-12,通过含毒介质菌碟法进行单因子试验及正交试验,对其发酵营养条件及培养条件进行了优化研究。试验结果表明:最佳培养基配方为玉米粉40 g/L,蔗糖40 g/L,蛋白胨3 g/L, KCl 0�5 g/L, MgSO4·7H2 O 0�5 g/L, K2 HPO41�0 g/L, FeSO4·7H2 O 0�01 g/L;最佳培养条件为装瓶量125 mL/250mL,初始pH为6�0,接种量6%,菌龄60 h,培养温度26℃,摇床转数175 r/min,发酵8 d。
