一株嗜酸氧化亚铁硫杆菌分离及生长特性研究

摘要

Acidithiobacillus ferrooxidan strain Y0611 is isolated from the acid mine drainage in a mine in Shanghang Fujian. A phylogenetic tree is constructed by comparing with the published 16S rDNA sequences of the relative bacterium species. In the phylogenetic tree, the Y0611 strain is the closest relative to some Acidithiobacillus ferrooxidans strains with more than 96 % sequence similarity. These results reveal that the Y0611 strain is a strain of ferrooxidans. The optimum growth conditions of Y0611 strain are determined as the optimum initial pH is 2.3, the best inoculation amount is 10 %, and culture temperature is about 30 °C.%从福建上杭某矿山酸性矿坑水中分离到一株嗜酸菌Y0611,以该菌16S rDNA同源性序列为基础构建了系统发育树,显示与多株嗜酸氧化亚铁硫杆菌16S rDNA序列有较高的同源性(＞96％),确定该菌为嗜酸氧化亚铁硫杆菌.确定了其生长最适条件,即在对数生长期,按10％接种量接种,培养液pH为2.3,30℃时振荡培养,Y0611菌生长活性最佳.
